[目的]寻找一种有效的泥火山土壤宏基因组DNA的提取方法。[方法]分别采用CTAB法、SDS加酶法、实验室改进法、试剂盒法提取泥火山土壤宏基因组,比较4种方法的提取效果。[结果]试剂盒法的提取效率最高、纯度最好;实验室改进法次之;SDS加酶法提取量最多。实验室改进法获得的DNA,稀释10倍可直接用于PCR扩增。[结论]从经济和DNA质量的角度考虑,实验室改进法可作为新疆泥火山土壤宏基因组DNA提取最有效的方法。
[Objective] The research aimed to find an effective extraction method of metagenomic DNA from mud volcano soil. [Methods] The macro-genomes of mud volcanoes soil were extracted by CTAB method, SDS-enzyme method, laboratory improvement method and kit method respectively. The extraction effects of four methods were compared. [Result] The kit method had the highest extraction efficiency, the highest purity, followed by the laboratory improvement method, and the SDS enzyme extraction method had the highest extraction rate. Laboratory improved DNA obtained, diluted 10 times can be directly used for PCR amplification. [Conclusion] From the perspective of economy and DNA quality, laboratory improvement method can be used as the most effective method for extracting genomic DNA from mud volcanoes in Xinjiang.
