目的:观察加味黑地黄丸对慢性支气管炎大鼠肺组织匀浆上清液中肿瘤坏死因子(TNF-α)、白细胞介素-8(IL-8)及其血清C反应蛋白(CRP)、可溶性细胞间黏附分子-1(sICAM-1)表达的影响。方法:将75只SD大鼠,用改良烟熏法复制慢性支气管炎大鼠模型,随机分为:模型对照组、地塞米松组、气管炎丸组、加味黑地黄丸(高、低剂量)组,另设正常对照组。灌胃给药20天。采用酶联免疫法(ELISA)测定各组大鼠肺组织匀浆中TNF-α和IL-8及其血清中CRP、sICAM-1的水平。结果:模型对照组大鼠肺组织匀浆上清TNF-α、IL-8和血清CRP、sICAM-1水平显著性升高(P<0.01)。高剂量加味黑地黄丸能降低大鼠肺组织匀浆上清TNF-α、IL-8和血清CRP、sICAM-1水平(P<0.01),与地塞米松作用基本一致;低剂量加味黑地黄丸仅降低大鼠血清sICAM-1水平(P<0.01)。结论:加味黑地黄丸能降低TNF-α、IL-8和CRP、sICAM-1的表达水平,是减轻慢性支气管炎大鼠气道炎症反应的有效机制之一。
Objective: To observe the effects of Jiawei Haodi Huangwan on the levels of tumor necrosis factor (TNF-α), interleukin-8 (IL-8) and serum C-reactive protein (CRP) in lung homogenate of chronic bronchitis rats, Effect of soluble intercellular adhesion molecule-1 (sICAM-1) expression. Methods: A total of 75 Sprague-Dawley rats were randomly divided into model control group, dexamethasone group, bronchitis pill group, Jiawei Haodi Huang Wan (high and low dose) Group, another set of normal control group. Gavage for 20 days. The levels of TNF-α and IL-8 in the lung homogenates and serum levels of CRP and sICAM-1 in each group were determined by enzyme-linked immunosorbent assay (ELISA). Results: The levels of TNF-α, IL-8 and serum CRP and sICAM-1 in the lung homogenate of model control group were significantly increased (P <0.01). High-dose flavonoids tablet could reduce TNF-α, IL-8 and serum CRP, sICAM-1 levels in lung homogenate of rats (P <0.01), which was consistent with the effect of dexamethasone; Pill only reduce serum sICAM-1 levels (P <0.01). CONCLUSION: Jiawei Hedihuang Pills can reduce the expression of TNF-α, IL-8, CRP and sICAM-1 and is one of the effective mechanisms to reduce airway inflammation in chronic bronchitis rats.
