目的建立高效液相色谱-电感耦合等离子体质谱联用技术(HPLC-ICP-MS)法测定单味雄黄和六神丸可溶性砷的含量方法;考察六神丸配伍对雄黄中可溶性砷溶出度的影响,探讨六神丸的配伍机制。方法分别建立了ICP-MS和HPLCICP-MS法测定雄黄及六神丸中总砷和可溶性砷含量的方法,采用人工胃液和人工肠液为溶出介质,考察雄黄和六神丸中可溶性砷的溶出情况,分析配伍使用对雄黄中可溶性砷溶出的影响。结果 5种不同形态砷标准物质在5~500μg·L-1内线性关系良好,精密度和重复性均良好,回收率89%~102%。可溶性砷从六神丸复方中的溶出量和溶出速率均明显低于从单味雄黄中的溶出。结论六神丸复方配伍可显著降低雄黄中可溶性砷的溶出,从而降低其毒性。
OBJECTIVE To establish a HPLC-ICP-MS method for the determination of soluble arsenic in single flavor realgar and Liushen Wan. To investigate the effect of compatibility of Liushen Wan on the solubility of soluble arsenic in realgar, The compatibility mechanism. Methods The contents of total arsenic and soluble arsenic in Realgar and Liushen Wan were determined by ICP-MS and HPLC-ICP-MS respectively. The dissolution of soluble arsenic in realgar and LiuShen Wan was studied by using artificial gastric juice and artificial intestinal juice as dissolution medium. Effect of Soluble Arsenic in Realgar. Results The results showed that the five standard forms of arsenic had a good linearity in the range of 5 ~ 500μg · L-1 with a good precision and reproducibility. The recoveries ranged from 89% to 102%. Soluble arsenic from Liushen Wan compound dissolution and dissolution rate were significantly lower than the single from realgar dissolution. Conclusion Liushen pill compound compatibility can significantly reduce the dissolution of realgar soluble arsenic, thereby reducing its toxicity.
