目的:研究不同原因所致心脏病患者机体内的骨髓间充质干细胞培养结果的差别。方法:分别采集并体外培养12例非冠心病和16例冠心病患者的骨髓间充质干细胞,观察其形态变化,对比分析其首次传代和每次传代时间、细胞增殖曲线和细胞表面标记的变化。结果:冠以病组中有4例无法体外培养出骨髓间充质干细胞。其余患者体外培养的骨髓间充质干细胞的形态无差异,均成梭形纤维状,早期呈克隆状生长;细胞的首次传代时间[冠心病组(16.7±7.2)d;非冠心病组(15.9±8.3)d]和生长曲线间也无明显差别;细胞表面标记分析显示CD54和CD106在非冠心病组的表达率分别为(19.08±0.97)%和(45.72±3.97)%,而它们在冠心病组的表达率则分别为(42.35±3.23)%和(65.90±4.33)%,非冠心病组明显低于冠心病组。结论:对于不同病因的心脏病患者,骨髓间充质干细胞在体外培养过程中其增殖上无明显差异,但是与功能相关的某些蛋白表达可能有所不同。
OBJECTIVE: To study the differences of the results of culturing bone marrow mesenchymal stem cells in heart patients with different causes. Methods: Bone marrow mesenchymal stem cells from 12 patients with non-coronary heart disease and 16 patients with coronary heart disease were collected and cultured in vitro respectively. Morphological changes were observed, and the changes of their primary passage, passage time, cell proliferation curve and cell surface markers . Results: There were 4 cases of bone marrow mesenchymal stem cells could not be cultured in vitro. The remaining patients had no morphological differences in the morphology of MSCs cultured in vitro, which were all spindle-shaped fibroblasts with early clonal growth. The passage time of the cells was 16.7 ± 7.2 d in coronary heart disease group and 15.9 ± 8.3) d] and the growth curve. The cell surface marker analysis showed that the expression rates of CD54 and CD106 in non-CHD group were (19.08 ± 0.97)% and (45.72 ± 3.97)%, respectively, The expression rates of heart disease group were (42.35 ± 3.23)% and (65.90 ± 4.33)%, respectively, which were significantly lower than those of non-coronary heart disease group. CONCLUSION: Bone marrow-derived mesenchymal stem cells have no significant difference in proliferation during the in vitro culture of heart disease patients with different etiologies, but the expression of certain protein related to function may be different.
