目的探讨孕酮及他莫西芬(tamoxifen,TAM)对子宫内膜癌细胞生长的影响及其对雌激素受体相关受体(ERRα)表达的影响。方法体外培养子宫内膜癌Ishikawa细胞,采用MTT法检测不同浓度孕酮及TAM对子宫内膜癌细胞生长的影响,同时采用Western Blot方法检测细胞中ERRα蛋白水平的变化。结果不同浓度孕酮对Ishikawa细胞体外增殖具有抑制作用,呈剂量-时间依赖性。TAM对Ishikawa细胞体外增殖具有双向作用,在×10-9~×10-7mol/L TAM作用下促进细胞增殖,并呈剂量-时间依赖性;在×10-5mol/L TAM作用下抑制细胞增殖。×10-9~10-6mol/L TAM作用后下调细胞中ERRα蛋白表达,×10-5mol/L TAM及孕酮作用后上调ERRα蛋白表达。结论孕酮及高浓度他莫西芬(×10-5mol/L)可抑制子宫内膜癌细胞的增殖,这种抑制作用可能通过调控细胞中ERRα表达实现;而低浓度他莫西芬则促进子宫内膜癌细胞的增殖。
Objective To investigate the effect of progesterone and tamoxifen (TAM) on the growth of endometrial cancer cells and its effect on the expression of estrogen receptor-related receptor (ERRα). Methods Ishikawa cells were cultured in vitro. MTT assay was used to detect the effects of different concentrations of progesterone and TAM on the growth of endometrial carcinoma cells. Meanwhile, the level of ERRα protein was detected by Western Blot. Results Different concentrations of progesterone inhibited the proliferation of Ishikawa cells in a dose-time-dependent manner. TAM had a bidirectional effect on Ishikawa cells proliferation in vitro, promoted cell proliferation under the dose of 10-9 ~ 10-7mol / L TAM in a time-and dose-dependent manner, and inhibited cell proliferation under the action of 10-5mol / L TAM . × 10-9 ~ 10-6mol / L TAM decreased ERRα protein expression in cells, × 10-5mol / L TAM and progesterone increased ERRα protein expression. Conclusion Progesterone and high concentrations of tamoxifen (10-5mol / L) can inhibit the proliferation of endometrial cancer cells, this inhibition may be through the regulation of ERRα expression in cells to achieve; and low concentrations of tamoxifen promote Endometrial cancer cell proliferation.
