目的:比较不同等级川芎饮片中藁本内酯和阿魏酸的含量,为传统川芎饮片分等方法提供理论依据。方法:采用高效液相色谱法测定藁本内酯、阿魏酸含量,色谱条件为Comatex C18色谱柱(4.6 mm×250 mm,5μm),柱温30℃,进样量10μL,流速1 mL·min-1;藁苯内酯、阿魏酸流动相分别为甲醇-水(70∶30),甲醇-1%乙酸溶液(30∶70),检测波长分别为326,321 nm。结果:一等、二等、统货川芎饮片中藁本内酯含量分别为3.14%,2.91%,0.30~2.84%;阿魏酸含量分别为0.26%,0.23%,0.1~0.21%。结论:川芎饮片中藁本内酯和阿魏酸含量为一等饮片最高,二等饮片次之,统货最低,证明川芎饮片的传统分级方法具有一定合理性。
OBJECTIVE: To compare the contents of ligustilide and ferulic acid in different grades of Chuanxiong decoction, providing the theoretical basis for the traditional grading method of Chuanxiong decoction. Methods: The ligustilide and ferulic acid were determined by high performance liquid chromatography. The chromatographic conditions were as follows: Comatex C18 column (4.6 mm × 250 mm, 5 μm), column temperature 30 ℃, injection volume 10 μL and flow rate 1 mL · min-1. The mobile phase of 藁Phenyllactone and ferulic acid were methanol-water (70:30) and methanol-1% acetic acid solution (30:70) respectively. The detection wavelength was 326,321 nm. Results: The contents of ligustilide in Chuanxiong Decoction were 3.14%, 2.91% and 0.30% ~ 2.84%, respectively. The contents of ferulic acid were 0.26%, 0.23% and 0.1 ~ 0.21% respectively. CONCLUSION: The contents of ligustilide and ferulic acid in Chuanxiong decoction are the highest, the second is the second, followed by the lowest, which proves that the traditional grading method of Chuanxiong decoction has certain rationality.
