背景:D-半乳糖老化小鼠脑内存在的β-淀粉样肽(β-amyloid,Aβ)产生过多,是否与脑老化过程有关?目的:观察中药救脑益智胶囊对D-半乳糖老化小鼠海马神经元Aβ及其相关蛋白表达的影响。设计:随机对照的实验研究。地点和对象:实验地点:首都医科大学宣武医院神经生化室。昆明小鼠60只,无特定病原体动物环境(SPF)级,雄性,体质量26~28g,由中国药品生物制品检定所实验动物中心提供。方法:采用免疫组织化学染色和免疫印记方法,检测D-半乳糖老化小鼠模型海马神经元Aβ、β-分泌酶、内质网Aβ结合蛋白(endoplasmicreticulumamyloidβ-peptidebindingprotein,ERAB)和早老蛋白-1的表达水平,并观察中药救脑益智胶囊的保护作用。主要观察指标:Aβ、β-分泌酶、ERAB和早老蛋白-1免疫组织化学染色结果和凝胶转移电泳分析。结果:对照组小鼠海马CA1区神经元有少量的Aβ及其相关蛋白表达,而D-半乳糖老化小鼠海马CA1区神经元Aβ及其相关蛋白表达水平明显增加,与对照组比较差异有高度显著性意义(P<0.01),中药组上述各种蛋白的表达结果与对照组接近。结论:D-半乳糖老化小鼠海马神经元存在着Aβ及其相关蛋白表达的上调,中药救脑益智胶囊可使之恢复正常水平,对神经元具有一定的营养和保护作用。
BACKGROUND: Excessive production of β-amyloid (Aβ) in the brain of D-galactose-aged mice has been associated with brain aging. Objective: To observe the effects of traditional Chinese medicine on brain-reducing glycosides for D-galactose Effects of aging mouse hippocampal neurons on the expression of Aβ and its related proteins . Design: Randomized controlled experimental study. Location and subject: Experimental site: Department of Neurochemistry, Xuanwu Hospital, Capital Medical University. There were 60 Kunming mice, no specific pathogenic animal environment (SPF) grade, male, and a body weight of 26-28 g, provided by the Experimental Animal Center of China Institute for the Control of Pharmaceutical and Biological Products. METHODS: Immunohistochemical staining and immunoblotting were used to detect Aβ, β-secretase, endoplasmic reticulum amyloid β-peptide binding protein (ERAB) and presenilin-1 in rat hippocampal neurons induced by D-galactose. The level of expression and observation of the protective effect of Chinese herbal rescue brain puzzle. MAIN OUTCOME MEASURES: Results of immunohistochemical staining for Aβ, β-secretase, ERAB, and presenilin-1, and gel electrophoresis analysis. RESULTS: A small amount of Aβ and its related proteins were expressed in hippocampal CA1 neurons of control mice, while the expression of Aβ and its related proteins in hippocampal CA1 neurons of D-galactose-treated mice was significantly increased. There was a difference compared with the control group. The significance was highly significant (P<0.01). The expression results of the various proteins in the TCM group were similar to those in the control group. Conclusion: There is an up-regulation of Aβ and its related proteins in hippocampal neurons of D-galactose-treated mice. The Chinese Herbal Rescue Syndrome can restore normal levels and has a certain nutritional and protective effect on neurons.
