应用Fura-2测定急性白血病(急白)骨髓或外周血中单个核细胞内钙离子浓度([Ca~(2+)]i)。肝素化血或骨髓液与RPMI1640混合后,滴入淋巴细胞分层液,离心。分离出的单个核细胞与Fura-2 AM,BSA,37℃温育40min。按激发波长350nm,发射波长500nm,测量其荧光强度F,Fmax,Fmin。用本法测定35例急白患者(51例次),结果表明[Ca~(2+)]i变化可作为一项反映化疗药物敏感性、预测化疗疗效的参考指标。
Fura-2 was used to determine the intracellular calcium concentration ([Ca ~ (2 +)] i) in mononuclear cells of acute leukemia (acute white) bone marrow or peripheral blood. Heparinized blood or bone marrow fluid and RPMI1640 mixed, drop into the lymphocyte stratification, and centrifuged. Isolated mononuclear cells were incubated with Fura-2 AM, BSA for 40 min at 37 ° C. Excitation wavelength 350nm, emission wavelength 500nm, measured fluorescence intensity F, Fmax, Fmin. 35 cases of acute white patients (51 cases) were determined by this method. The results showed that the change of [Ca ~ (2 +)] i could be used as a reference index to reflect the sensitivity of chemotherapy drugs and predict the efficacy of chemotherapy.
