为了发掘控制红掌(Anthurium andraeanum)主要观赏器官——佛焰苞发育的关键基因类型,本研究克隆的一个红掌B功能MADS-box基因。通过序列比对和结构域分析,发现该基因属于PI亚家族,命名为AaPI。生物信息学分析结果显示,该基因在氨基酸组成上高度保守,进化树分析认为该红掌基因处于单子叶与双子叶植物交界处,暗示该物种在分子进化上较为原始。半定量和荧光定量RT-PCR结果显示该基因在红掌生殖器官大量表达,苞片中的表达水平仅次于花序,而在其营养器官中则表达很少或不表达。本研究为丰富在分子水平上对红掌器官发生的认识做了积极探索。
In order to find out the key gene types controlling the development of spathes, which is the main ornamental organ of anthurium andraeanum, we cloned a anthracnose B-functional MADS-box gene. Through sequence alignment and domain analysis, it was found that the gene belongs to PI subfamily named AaPI. Bioinformatics analysis showed that the gene was highly conserved in amino acid composition. The phylogenetic tree analysis indicated that this gene is located at the junction of monocotyledon and dicotyledon, indicating that the species is more primitive in molecular evolution. Semi-quantitative and fluorescent quantitative RT-PCR results showed that the gene was abundantly expressed in the genus Anthurium, the expression level in bracts was second only to inflorescences, and the expression was little or not expressed in vegetative organs. This study actively explores the understanding of the occurrence of Anthurium by molecular level.
