目的:采用HPLC法建立亚麻叶中绿原酸、荭草素、异荭草素、牡荆素、异牡荆素含量的测定方法。方法:Inert Sustain C18(250 mm×4.6 mm,5μm)色谱柱;流动相为乙腈(A)-0.1%甲酸溶液(B),梯度洗脱;柱温为40℃;流速为1.0 m L/min;进样量为5μL;检测波长为350 nm。结果:绿原酸、荭草素、异荭草素、牡荆素、异牡荆素分别在4.00~40.02μg/m L(r1=0.9998)、3.44~34.44μg/m L(r2=0.9999)、3.53~35.34μg/m L(r3=0.9999)、2.55~25.50μg/m L(r4=0.9999)、4.33~43.32μg/m L(r5=0.9998)范围内呈良好线性关系;平均加样回收率分别为100.01%(RSD为0.64%)、98.85%(RSD为1.13%)、98.65%(RSD为2.62%)、98.29%(RSD为2.34%)、103.06%(RSD为3.38%)。结论:该方法简便,结果准确、可靠、重复性好,可用于测定亚麻叶中这5种成分的含量。
OBJECTIVE: To establish a method for the determination of chlorogenic acid, turmeric, isohumulone, vitexin and isovitaminin in flax leaves by HPLC. Methods: Inert Sustain C18 (250 mm × 4.6 mm, 5 μm) column was used. The mobile phase consisted of acetonitrile (A) -0.1% formic acid solution (B) with gradient elution. The column temperature was 40 ℃ and the flow rate was 1.0 m L / min ; Injection volume of 5μL; detection wavelength of 350 nm. Results: The concentrations of chlorogenic acid, toardin, isoorientin, vitexin and isovitamin were 4.00-40.02μg / mL (r1 = 0.9998) and 3.44-34.44μg / mL (r2 = 0.9999) , 3.53-35.34μg / m L (r3 = 0.9999), 2.55-25.50μg / m L (r4 = 0.9999), 4.33-43.32μg / m L (r5 = 0.9998) Rates were 100.01% (RSD 0.64%), 98.85% (RSD 1.13%), 98.65% (RSD 2.62%), 98.29% (RSD 2.34%) and 103.06% (RSD 3.38%), respectively. Conclusion: The method is simple, accurate, reliable and reproducible. It can be used to determine the contents of these five components in flax leaves.
