目的类风湿性关节炎(RA)是常见的系统性免疫性疾病,本研究探讨色胺酰-tRNA合成酶(TTS)在RA患者外周血中的表达,探讨其与RA疾病活动度间的关联。方法应用荧光定量PCR法检测68例RA患者、68例健康人PBMCs上TTS的mRNA表达,探讨其与IDO、Foxp3和RoR-γt mRNA间的相关性,并研究其与疾病活动度间的关系。结果 RA患者外周血TTS mRNA表达显著高于对照组,差异有统计学意义(P<0.01),且与ESR、DAS28呈正相关(P<0.01)。而RA患者外周血IDO、Foxp3和RoR-γt mRNA表达差异无统计学意义,且与TTS mRNA无相关性(P>0.05)。结论外周血PBMCs中TTS mRNA表达异常升高可引起RA患者色氨酸代谢失调,可能在RA慢性炎症过程中发挥免疫调节作用。
Objective Rheumatoid arthritis (RA) is a common systemic immune disease. In this study, the expression of TTS in the peripheral blood of patients with RA was studied to explore its association with the activity of RA disease . Methods Fluorescence quantitative PCR was used to detect the mRNA expression of TTS in PBMCs from 68 patients with RA and 68 healthy individuals. The correlation between TTS mRNA and IDO, Foxp3 and RoR-γt mRNA was analyzed and the relationship between TTS and disease activity was also studied. Results The expression of TTS mRNA in peripheral blood of patients with RA was significantly higher than that of the control group (P <0.01), and positively correlated with ESR and DAS28 (P <0.01). However, the expression of IDO, Foxp3 and RoR-γt in peripheral blood of RA patients showed no significant difference (P> 0.05). Conclusion The abnormal expression of TTS mRNA in peripheral blood PBMCs may cause the abnormal metabolism of tryptophan in RA patients and may play an immunomodulatory role in the process of RA chronic inflammation.
