目的:探讨肺小细胞癌( S C L C) 的p53 基因改变与mdm2 基因蛋白表达的关系。方法:应用免疫组化 L S A B 法和聚合酶链反应单链构象多态性( P C R S S C P) 分析的方法,对14 例 S C L C 的石蜡切片组织进行研究。结果:免疫组化染色p53 蛋白阳性9 例,阳性率为643 % (9/14) ;mdm2 蛋白阳性4 例,阳性率为286 % (4/14) 。p53 基因第5 、6 、7 、8 外显子,突变率分别为214 % (3/14) ;143(2/14) ;143 % (2/14) ;71 % (1/14) ,总突变率为571 % (8/14) 。结论: S C L C 中存在较高的p53 基因突变率和mdm2 基因蛋白的明显表达。
Objective: To investigate the relationship between p53 gene mutation and mdm2 gene protein expression in small cell lung cancer (S C L C). METHODS: Paraffin-embedded tissues of 14 cases of S C L C were studied by immunohistochemical L S A B method and polymerase chain reaction single-strand conformation polymorphism (P C R S S C P) method. . Results: The immunohistochemical staining of p53 protein was positive in 9 cases, the positive rate was 64. 3% (9/14); mdm 2 protein was positive in 4 cases, the positive rate was 28.6% (4/14). Exon 5, 6, 7, and 8 of p53 gene had mutation rates of 214 % (3/14); 143(2/14); 143 % (2/14); 71. % (1/14), the total mutation rate was 57.1% (8/14). Conclusion: There is a high mutation rate of p53 gene and obvious expression of mdm2 gene protein in S C L C .
