目的:分析埃兹蛋白(ezrin)与CD44在肾细胞癌(renal cell carcinoma,RCC)中的表达情况及其临床意义。方法:选取2012年2月至2012年11月湖南省肿瘤医院泌尿外科收治的56例RCC患者的癌组织石蜡切片标本及17例癌旁组织标本,采用免疫组化技术检测标本中ezrin与CD44的表达情况,并分析两者表达的相关性及与RCC各临床病理指标的关系。结果:RCC中埃兹蛋白[60.7%(34/56)vs 29.4%(5/17),P<0.05]与CD44蛋白[64.3%(36/56)vs 0(0/17),P<0.01]表达水平明显高于癌旁组织。结合病理诊断结果研究发现,CD44蛋白和埃兹蛋白表达水平随RCC病理分级、临床分期的升高而升高(P<0.05);在肿瘤最大径>7.0 cm的RCC组织中,埃兹蛋白的阳性表达率显著高于最大径≤7.0 cm的RCC组织(P<0.05),而CD44阳性表达率无显著差异(P>0.05)。不同性别的患者组织内埃兹蛋白、CD44的表达水平无显著差异(P>0.05)。Spearman分析结果表明,CD44与埃兹蛋白在RCC组织中的表达水平呈高度正相关(r=0.427,P<0.01)。结论:埃兹蛋白与CD44在RCC组织中均呈高表达,两者表达呈正相关;埃兹蛋白与CD44在RCC的发生发展及浸润转移过程中扮演着重要角色。
Objective: To analyze the expression of ezrin and CD44 in renal cell carcinoma (RCC) and its clinical significance. Methods: A total of 56 specimens of RCC from paraffin-embedded sections from Hunan Provincial Tumor Hospital from February 2012 to November 2012 were collected for paraffin section and para-cancerous tissues from 17 cases. Immunohistochemistry was used to detect the expression of ezrin and CD44 Expression, and analyze the correlation between the expression and the relationship with the clinicopathological parameters of RCC. Results: Compared with CD44 protein [64.3% (36/56) vs 0 (0/17), P <0.01), the percentage of ezrin protein in RCC [60.7% (34/56) vs 29.4% ] Expression was significantly higher than adjacent tissues. According to the results of pathological diagnosis, the expression of CD44 protein and Ezrin protein increased with the increase of RCC grade and clinical stage (P <0.05). In RCC tissues with the largest diameter> 7.0 cm, the expression of Ezrin (P <0.05), while the positive rate of CD44 was not significantly different (P> 0.05). There was no significant difference in the expression levels of Ezrin and CD44 between different sexes (P> 0.05). Spearman analysis showed that there was a highly positive correlation between CD44 and Ezrin expression in RCC tissues (r = 0.427, P <0.01). CONCLUSION: Ezrin and CD44 are highly expressed in RCC tissues, and their expressions are positively correlated. Ezrin and CD44 play an important role in the development of RCC and invasion and metastasis.
