目的:研究人参果总皂苷对体外培养的肿瘤细胞作用,并初步探讨其作用机制。方法:MTT法测定人参果总皂苷对人宫颈癌细胞(Hela)细胞株的增殖的影响;电镜观察人参果总皂苷对肿瘤细胞形态学的影响;Western blot免疫印记法检测人参果总皂苷对Hela细胞细胞周期相关蛋白表达的影响。结果:0.125、0.25、0.50、1.00、2.00、4.00μg/ml人参果总皂苷对Hela细胞的增殖均有抑制作用;0.5μg/ml人参果总皂苷作用于Hela细胞后,透射电镜下可见细胞核染色质边集、核碎裂及凋亡小体情况,具有一定的促进肿瘤细胞凋亡的作用;cyclin和CDK4的蛋白表达逐渐减少。结论:人参果总皂苷对肿瘤细胞的增殖具有抑制作用,其作用机制可能与促进肿瘤细胞的凋亡有关诱导其凋亡使细胞停滞于G2/M期密切相关。
Objective: To study the effect of ginseng fruit saponins on tumor cells in vitro and to explore its mechanism. Methods: MTT assay was used to determine the effect of total ginsenosides on the proliferation of Hela cells. The mechanism may be related to the promotion of apoptosis of tumor cells and the induction of apoptosis so that cells arrest in G2 / M phase.
