二甲砷基谷胱甘肽是由二甲基砷与谷胱甘肽结合而成的新型有机砷化合物,在体内外对多种血液系统肿瘤及实体瘤敏感。其作用机制包括针对性地破坏癌细胞线粒体的功能,影响癌细胞信号转导,阻止癌细胞的血管再生,从而导致癌细胞的生长周期停滞和细胞凋亡。二甲砷基谷胱甘肽导致癌细胞凋亡的机制不同于三氧化二砷,因此能杀灭对三氧化二砷具有耐药性的癌细胞。本文对其治疗作用机制、药理学、药动学研究及安全性等作一综述。
Dimethylarsin glutathione is a new organic arsenic compound formed by the combination of dimethyl arsenic and glutathione, which is sensitive to various hematological tumors and solid tumors both in vitro and in vivo. Its mechanism of action includes targeted destruction of mitochondrial function of cancer cells, affecting cancer cell signal transduction, preventing the revascularization of cancer cells, resulting in cancer cell cycle arrest and apoptosis. The mechanism by which arsenomethionine causes apoptosis in cancer cells is different from that of arsenic trioxide and thus can kill cancer cells that are resistant to arsenic trioxide. This article reviews its mechanism of action, pharmacology, pharmacokinetics and safety.
