目的:研究金匮肾气丸含药血清对体外兔骨髓间充质干细胞(bone marrow mesenchymal stem cells BMSCs)增殖,细胞周期及凋亡的影响。方法:使用全骨髓贴壁法分离兔BMSCs,通过MTT法检测金匮肾气丸含药血清对BMSCs活性的影响,通过流式细胞仪(Fow Cytometry FCM)检测金匮肾气丸含药血清对地塞米松(dexamethasone DEX)诱导的BMSCs的凋亡率和细胞周期的影响。结果:金匮肾气丸各组光吸收值(OD)高于空白对照组。金匮肾气丸各组与模型组比较,Go/G1期细胞比例降低,S期和G2/M期细胞比例升高,凋亡率降低,并呈一定的量效依赖关系。结论:金匮肾气丸含药血清体外能够提高兔BMSCs的活性,促进G0/G1期的BMSCs向S期和G2/M期转化,促进BMSCs的增殖,降低地塞米松所致的兔BMSCs的凋亡率。
Objective: To study the effect of Jinkuishenqiwan containing serum on proliferation, cell cycle and apoptosis of rabbit bone marrow mesenchymal stem cells (BMSCs) in vitro. METHODS: Rabbit BMSCs were isolated by whole bone marrow adherence method. The effect of Jingui Shenqi pill containing serum on the activity of BMSCs was detected by MTT assay. The effect of Jingui Shenqi Pill on serum BMSCs was assayed by flow cytometry (Fow Cytometry FCM) Effect of dexamethasone DEX on apoptosis rate and cell cycle of BMSCs. Results: The light absorption value (OD) of Jinkuishenqiwan group was higher than that of the blank control group. Compared with the model group, the proportion of cells in Go / G1 phase was decreased, the proportion of cells in S phase and G2 / M phase was increased, the apoptosis rate was decreased, and there was a certain dose-effect relationship. CONCLUSION: Jinkuishenqiwan serum can increase the activity of BMSCs in vitro and promote the transformation of BMSCs into S phase and G2 / M phase in G0 / G1 phase, promote the proliferation of BMSCs and decrease the BMSCs dexamethasone-induced Apoptosis rate.
