[目的]观察二硫化碳(CS_2)对雄性大鼠睾丸组织病理改变及细胞凋亡的影响。[方法]24只健康雄性Wistar大鼠随机分为4组,以不同浓度CS_2(0、50、250、1250mg/m~3)吸入染毒,每天2 h,每周5 d,共10周。染毒结束后,取大鼠睾丸以HE染色,采用普通光镜、电镜及末端转移酶标记技术(TUNEL法)观察在CS_2作用下,睾丸组织的病理变化及细胞凋亡的情况。[结果]随CS_2染毒浓度的增加,大鼠睾丸间质水肿加重;高浓度组曲细精管生精细胞数目及层次明显减少。在低、中浓度组可见间质细胞内质网扩张,精原细胞、初级精母细胞的线粒体发生肿胀;高浓度组支持细胞、间质细胞、精原细胞、初级精母细胞及精子细胞均出现胞质空洞,精原细胞、初级精母细胞线粒体肿胀加重,精子细胞线粒体也出现肿胀,精子纵切面“9*2+2”轴丝结构紊乱,微管溶解缺失。随CS_2染毒浓度的增加,睾丸组织中细胞凋亡指数增高,中、高浓度组细胞凋亡指数与对照组比较有统计学意义(P<0.05),异常增多的凋亡细胞以精母细胞为主。[结论]CS_2可导致大鼠睾丸组织发生病理变化,生精细胞异常凋亡。
[Objective] To observe the effect of carbon disulfide (CS_2) on pathological changes and apoptosis of testis in male rats. [Methods] Twenty-four healthy male Wistar rats were randomly divided into 4 groups. The rats were exposed to different concentrations of CS 2 (0, 50, 250 and 1250 mg / m 3) for 2 days and 5 days a week for 10 weeks. After the exposure, the testis of rats was stained with hematoxylin and eosin (HE), and the pathological changes and apoptosis of testis were observed under ordinary light, electron microscope and TUNEL. [Result] The testicular interstitial edema was aggravated with the increase of CS 2 concentration. The number and level of spermatogenic cells in seminiferous tubules in high concentration group were significantly decreased. The mitochondria of spermatogonia and primary spermatocyte were swollen in the medium and low concentration groups, and the mitochondria in spermatogonia and primary spermatocytes were swollen. In the high concentration group, the number of supporting cells, interstitial cells, spermatogonia, primary spermatocytes and spermatids Cytoplasmic cavity, spermatogonia, primary spermatocyte mitochondrial swelling increased sperm cell mitochondria also swollen, sperm longitudinal section “9 * 2 +2 ” axial filament disorder, lack of microtubule lysis. With the increase of CS 2 concentration, the apoptosis index of testes increased, and the apoptosis index of medium and high concentration groups was statistically significant compared with that of control group (P <0.05). The abnormally increased apoptotic cells were expressed as spermatocytes Mainly. [Conclusion] CS 2 can induce pathological changes in testicular tissue of rats and abnormal apoptosis of spermatogenic cells.
