目的:利用响应面分析法对鬼针草叶总黄酮纯化工艺进行优化。方法:以芦丁的含量为指标,考察上样流速、上样浓度、p H值、洗脱剂浓度、柱子径高比等因素,通过Box-Behnken设计建立响应面模型来优选鬼针草叶中总黄酮的纯化工艺参数。结果:选用HPD-100型大孔吸附树脂,上样流速为2 BV/h,p H为6,上样浓度为2.027 mg/m L,70%乙醇洗脱,柱子径高比为1∶15。结论:该方法纯化工艺良好,总黄酮的纯度可达到62.86%,与理论值61.88%的相对误差为1.56%
OBJECTIVE: To optimize the purification of total flavonoids from Bidens bipinnata by response surface methodology. Methods: Using the content of rutin as an index, the response surface model was designed and established by Box-Behnken to optimize the parameters of flow rate, sample concentration, p H value, eluent concentration and column diameter to height ratio. Purification of total flavonoids in the process parameters. Results: HPD-100 macroporous resin was used. The sample flow rate was 2 BV / h, pH was 6, sample concentration was 2.027 mg / mL and 70% ethanol was eluted. The column height-diameter ratio was 1:15 . Conclusion: The purification process of the method is good, the purity of total flavonoids can reach 62.86%, the relative error with the theoretical value of 61.88% is 1.56%
