目的通过比较整合素aV及血管内皮生长因子(VEGF)在人脑海绵状血管瘤和正常脑组织血管标本中的表达情况,探讨脑海绵状血管瘤发生和发展的可能机制。方法采集人脑海绵状血管瘤标本40例及对照血管组织10例,扩增标本中整合素aV和VEGF的对应mRNA序列,观察两种标本血管成分中整合素aV和VEGF蛋白表达情况,比较其相对含量及两者相关性。结果 40例试验标本中VEGF表达均为阳性,具有显著性差异(P=0.001),整合素aV在脑海绵状血管瘤中的表达明显高于脑动静脉畸形和对照血管。结论脑海绵状血管瘤中的血管生长可能与整合素aV相关。海绵状血管瘤的形成、生长和破裂可能与其EC中VEGF的高表达有关。
Objective To investigate the possible mechanism of the occurrence and development of cavernous hemangiomas by comparing the expression of integrin aV and vascular endothelial growth factor (VEGF) in human cavernous hemangiomas and normal brain blood vessels. Methods Forty cases of human cavernous hemangiomas and 10 cases of control vascular tissues were collected. The corresponding mRNA sequences of integrin aV and VEGF were amplified, and the expression of integrin aV and VEGF in two samples were observed. Relative content and their correlation. Results The expression of VEGF in all the 40 specimens was positive, with significant difference (P = 0.001). The expression of integrin aV was significantly higher in cerebral cavernous hemangiomas than in cerebral arteriovenous malformations and control vessels. Conclusion Angiogenesis in cavernous hemangiomas may be related to integrin aV. Cavernous hemangioma formation, growth and rupture may be related to its high expression of VEGF in the EC.
