目的目的观察红花对结扎单侧输尿管诱导的肾间质纤维化实验大鼠肾小管上皮细胞表型转化的抑制作用。方法结扎单侧输尿管方式复制肾间质纤维化实验动物模型,分别给以红花4.0g·kg-1·d-1、缬沙坦10mg·kg-1·d-1经口灌服,采用免疫组化、原位杂交方法检测大鼠肾脏肾小管上皮细胞表型转化标志物α-平滑肌肌动蛋白(α-SMA)及Ⅲ型胶原的表达。结果单侧输尿管结扎大鼠肾脏α-SMA动蛋白和Ⅲ型胶原的阳性面积及积分光密度较假手术组明显增强,红花及缬沙坦可显著性抑制其表达。结论红花可以通过抑制肾小管上皮细胞的表型转化拮抗肾间质纤维化。
Objective To observe the inhibitory effect of safflower on the phenotypic transformation of renal tubular epithelial cells induced by ligation of unilateral ureteral obstruction in rats with renal interstitial fibrosis. Methods Rat model of renal interstitial fibrosis was established by ligation of unilateral ureteral obstruction. The rats were orally administrated with safflower 4.0g · kg-1 · d-1 and valsartan 10mg · kg-1 · d-1 respectively. Immunohistochemistry and in situ hybridization were used to detect the expression of α-smooth muscle actin (α-SMA) and type Ⅲ collagen in rat renal tubular epithelial cells. Results The positive area and integral optical density of α-SMA and Ⅲ collagen in the kidney of unilateral ureteral obstruction group were significantly increased than those in sham operation group, and safflower and valsartan significantly inhibited the expression of α-SMA and Ⅲ. Conclusion Safflower can antagonize renal interstitial fibrosis by inhibiting the phenotypic transformation of renal tubular epithelial cells.
