目的:研究不同浓度二根龙蛭汤浓缩煎剂的主要药用化学成分含量。方法:将同剂量二根龙蛭药方分成1组和2组,分别加入100mL、200mL水进行煎煮,过滤后采用溶剂法、色谱法进行分离、鉴别,采用高效液相色谱法测定不同制备方法制得的二根龙蛭汤浓缩煎剂中药用化学成分水蛭素、丹参酮Ⅱ的含量。结果:1组煎剂消耗凝血酶的体积(14.56U)明显高于2组(11.52),表明1组煎剂中水蛭素的含量高于2组中水蛭素的含量;此外,1组煎剂中丹参酮ⅡA的含量(0.098 73)也高于2组(0.098 50)。结论:采用100mL水煎二根龙蛭汤所得药液所含水蛭素和丹参酮ⅡA比200mL水煎液的含量高,药效更佳。
Objective: To study the main medicinal contents of two decoctions of Longzhi Decoction at different concentrations. Methods: Divide the two doses of Radix leprosi with the same dose into 1 group and 2 groups, respectively, add 100mL, 200mL water for decoction, and then separate and identify by filtration and separation by solvent method and HPLC method. The different preparation methods were determined by high performance liquid chromatography Preparation of the two decoction Long Zhi Tang decoction of the chemical constituents hirudin, tanshinone Ⅱ content. Results: The volume of thrombin consumed by 1 decoction group (14.56U) was significantly higher than that of 2 groups (11.52), indicating that the content of hirudin in 1 decoction group was higher than that in 2 groups. In addition, The content of tanshinone IIA (0.098 73) was also higher than that of the two groups (0.098 50). Conclusion: The content of hirudin and tanshinone ⅡA contained in 100mL decoction of Radix et Rhizoma Rhei decoction is higher than that of 200mL decoction, and the efficacy is better.
