为建立裸小鼠皮下人脑胶质瘤模型,体外培养SHG44人脑胶质瘤细胞,用无血清1640培养液洗涤和离心两次,行活细胞计数后以甲氧氟烷吸入方式麻醉裸小鼠,分别将50μl含1,2,3×10~6个SHG44细胞的无血清悬液注入各组裸小鼠右腿上部皮下,现察SHG44细胞裸小鼠皮下致瘤性并计算肿瘤大小。30d后处死裸小鼠,取其肝、肺和皮下肿瘤组织,进行HE染色和电镜检查,确定肿瘤本质和有无肿瘤转移,比较各组裸小鼠肿瘤生长特点。研究结果表明建立裸鼠皮下人脑胶质瘤模型,以2×10~6个SHG44细胞接种效果较理想,未发现肿瘤远处转移。所建裸小鼠皮下人脑胶质瘤模型为人脑胶质瘤化疗、放疗和基因治疗等相关研究奠定了基础。
To establish a model of subcutaneous human glioma in nude mice, SHG44 human glioma cells were cultured in vitro and washed twice with serum-free 1640 medium and centrifuged twice with viable cell count, followed by metoprolol inhalation Mice were injected with 50μl serum-free suspension containing 1, 2, 3 × 10-6 SHG44 cells into the subcutaneous of the upper right leg of each group. The subcutaneous tumorigenicity of SHG44 cells was observed and the tumor size was calculated. After 30 days, the nude mice were sacrificed and their liver, lung and subcutaneous tumor tissues were harvested for HE staining and electron microscopy to determine the nature of the tumor and whether there was tumor metastasis. The tumor growth characteristics of the nude mice in each group were compared. The results showed that the establishment of subcutaneous human glioma model in nude mice, 2 × 10 ~ 6 SHG44 cell vaccination effect is satisfactory, no distant metastasis was found. The nude mouse model of subcutaneous human glioma laid the foundation for the related research of human glioma chemotherapy, radiotherapy and gene therapy.
