目的 探讨局部照射对创伤愈合过程中伤口 (Fb)TGFβ1及TGFβR1表达的影响及意义。方法 将 1 32只Wistar大鼠分为单纯创伤组 (对照组 )和照射复合创伤组 (照射组 ) ,用免疫组织化学、原位杂交和图像分析方法检测两组伤口愈合过程中伤后 1~ 60d各时间点伤口成纤维细胞中TGFβ1、TGFβR1蛋白及TGFβ1mRNA表达的变化规律。 结果 单纯创伤组伤口成纤维细胞TGFβ1、TGFβR1表达有明显的时相性 ,伤后 2~ 2 1d可见表达 ,1 0d为表达高峰 ;照射复合创伤组其表达减弱 ,且表达时相延长 ,伤后 2 8d仍有表达。结论 照射抑制伤口成纤维细胞内源性TGFβ1及TGFβR1的表达 ,表明照射使成纤维细胞自分泌TGFβ1功能减弱并对外源性TGFβ1的反应性减弱
Objective To investigate the effect and significance of local irradiation on the expression of TGFβ1 and TGFβR1 in wounds (Fb) during wound healing. Methods One hundred and thirty-two Wistar rats were divided into simple wound group (control group) and irradiated composite wound group (irradiation group). Immunohistochemistry, in situ hybridization and image analysis were used to detect the wound healing in both groups during 1 ~ 60 days The changes of TGFβ1, TGFβR1 protein and TGFβ1 mRNA expression in wound fibroblasts at different time points. Results The expression of TGFβ1 and TGFβR1 in wounds of wounds in simple wound group were obviously time-phased. The expression of TGFβ1 and TGFβR1 was observed 2 ~ 21 days after wounding and peaked at 10 days. The expression of TGFβ1 and TGFβR1 was attenuated and the expression of TGFβ1 was prolonged 8d still expressed. Conclusions Irradiation inhibits the expression of endogenous TGFβ1 and TGFβR1 in wound fibroblasts, indicating that irradiation attenuates the autocrine function of TGFβ1 and decreases the reactivity to exogenous TGFβ1
