目的 探讨细胞周期蛋白B1(cyclinB1)在白血病发生、发展中的作用及对成人急性白血病 (AL)预后的判断价值。方法 对 85例成人AL初治患者 ,并对其中 4 7例达到缓解及 18例复发的患者、10例持续完全缓解 (CCR)期的AL患者采用流式细胞术检测cyclinB1、p2 1cip1蛋白表达水平和细胞周期分布 ;采用半定量RT PCR检测cyclinB1、p2 1cip1、增殖细胞核抗原 (PCNA)mRNA水平。结果 85例初治AL患者全周期或G1/S期cyclinB1蛋白表达水平明显高于正常对照 (P <0 .0 0 1) ;缓解患者cy clinB1蛋白表达与正常对照组比较差异无统计学意义 (P =0 .2 1) ,复发患者cyclinB1蛋白表达高于正常对照 (P <0 .0 5 ) ,但低于AL初治组 (P <0 .0 1) ,CCR组cyclinB1与正常对照组比较差异无显著性 (P=0 .6 94 ) ;初治AL患者的cyclinB1表达率高于正常对照组 (P <0 .0 0 1) ;cyclinB1蛋白高表达者均有G1期异常高表达 ;cyclinB1与p2 1的蛋白表达呈负相关 ;cyclinB1蛋白表达与其mRNA水平呈正相关 ;cy clinB1表达与细胞增殖指数 (PI)及PCNA表达水平呈正相关 ;cyclinB1表达水平高的患者有较高的治疗缓解率 (P <0 .0 1) ,但有较高的复发率 (P <0 .0 1) ,cyclinB1高表达者有较高的生存率。结论 cyclinB1在成人AL患者中有过度表达和周期分布 ,其
Objective To investigate the role of cyclinB1 in the development and progression of leukemia and its prognostic value in adult acute leukemia (AL). Methods A total of 85 adult patients with AL-naive disease were enrolled in this study. Forty-seven patients with remission and 18 relapsed patients were enrolled in this study. Flow cytometry was used to detect the expression of cyclinB1 and p2 1cip1 protein in 10 AL patients with complete remission (CCR) And cell cycle distribution. The mRNA expressions of cyclinB1, p2 1cip1 and PCNA were detected by semi-quantitative RT-PCR. Results The expression of cyclinB1 protein in 85 patients with initially untreated AL was significantly higher than that in the normal controls (P <0.01). There was no significant difference in the expression of cy clinB1 protein between the two groups (P> 0.05) P = 0.21). The expression of cyclinB1 in relapsed patients was significantly higher than that in normal control group (P <0.05), but lower than that in AL group (P <0.01). The cyclinB1 in CCR group was significantly higher than that in normal control group The difference was not significant (P = 0.644). The expression of cyclinB1 in newly diagnosed AL patients was higher than that in normal control group (P <0.01). The high expression of cyclinB1 protein in G1 phase was abnormally high expression. CyclinB1 And cyclinB1 protein expression was positively correlated with its mRNA level; cy clinB1 expression and cell proliferation index (PI) and PCNA expression was positively correlated; high cyclinB1 expression in patients with high response rate ( P <0.01), but there was a higher recurrence rate (P <0.01). Patients with high expression of cyclinB1 had a higher survival rate. Conclusion CyclinB1 is over-expressed and periodically distributed in adult AL patients
