目的探讨大鼠脑缺血后,干扰素-γ(IFN-γ)是否参与免疫损伤和IFN-γ在脑缺血中的作用。方法用线拴封闭大脑中动脉的方法制作脑缺血动物模型,采用原位杂交的方法检测缺血脑组织IFN-γmRNA表达动态的变化。结果(1)缺血脑半球和假手术的脑半球相比较,在1h时IFN-γmRNA的数量就显著增加(P<0.001),且随着脑缺血时间延长IFN-γ表达量增加,6hP<0.001,12hP<0.01,24hP<0.001,48hP<0.001,6dP<0.001。(2)IFN-γmRNA表达数量损伤面积扩大而增加;结论在脑缺血过程中IFN-γmRNA表达增加,IFN-γmRNA表达主要参与大脑损伤后期反应过程。
Objective To investigate whether interferon-γ (IFN-γ) is involved in immune injury and IFN-γ in cerebral ischemia in rats after cerebral ischemia. Methods The cerebral ischemic model was established by occluding the middle cerebral artery with a tethered cord. The dynamic changes of IFN-γ mRNA in ischemic brain tissue were detected by in situ hybridization. Results (1) The numbers of IFN-γmRNA in ischemic cerebral hemisphere and sham-operated cerebral hemisphere were significantly increased at 1h (P <0.001), and the expression of IFN-γ increased with the prolongation of cerebral ischemia. <0.001, 12 hP <0.01, 24 hP <0.001, 48 hP <0.001, 6 dP <0.001. (2) The number of IFN-γmRNA expression increased with the enlargement of the area. Conclusion The expression of IFN-γmRNA increased during the process of cerebral ischemia. The expression of IFN-γmRNA mainly involved in the process of brain injury.
