以兔眼蓝莓‘顶峰’的带芽茎段为外植体,研究了腋芽诱导、增殖、生根和出瓶移栽的最佳培养条件,建立了‘顶峰’的离体快繁技术体系。结果表明:适宜芽诱导的培养基为WPM+ZT0.5mg/L;适宜苗增殖的培养基为WPM+ZT 1mg/L;适宜组培苗生根的培养基为WPM+ZT1.0mg/L+IBA 0.5mg/L。在生根培养基中用河沙代替琼脂粉固定植株,易于清洗出瓶苗根系上粘附的培养液,避免损伤不定根系,60d后组培苗的移栽成活率达80%。
The best culture conditions for inducing, multiplying, rooting and transplanting of axillary buds were studied with stem buds with the peak of blueberry in rabbit eyes as explants. The results showed that the optimum culture medium was WPM + ZT0.5mg / L; the suitable culture medium for proliferation was WPM + ZT1mg / L; the suitable culture medium for rooting culture was WPM + ZT1.0mg / L + IBA 0.5mg / L. In the rooting medium, river sand was used instead of agar to fix the plants. It is easy to wash out the adherent culture solution on the root of the bottle seedling to avoid adventitious root injury. After 60 days, the transplanting survival rate of the tissue culture seedlings reached 80%.
