目的:观察活血通督汤对急性脊髓损伤后神经生长因子(NGF)和胶质纤维酸性蛋白(GFAP)表达的影响,探讨活血通督汤对脊髓损伤干扰的作用机制。方法:将24只成年雌性SD大鼠随机分为假手术组、模型组和活血通督汤组,每组8只。术前、术后7d均予连续灌胃,活血通督汤组灌服活血通督汤,另两组灌服等量0.9%氯化钠溶液,均为每日2次。假手术组椎板摘除暴露脊髓但不予打击,模型组和活血通督汤组采用改良Allen’s法制备脊髓损伤模型。术后7d取材,灌注固定、石蜡包埋。采用尼氏染色法观察脊髓及神经元形态变化,免疫荧光法检测脊髓中NGF和GFAP的表达情况。结果:假手术组脊髓灰质呈蝴蝶状,结构完整,灰白质界限清楚,胞质内可见虎斑样尼氏体;与模型组比较,活血通督组瘀血面积较小,神经元水肿较轻,神经元形态较好,未见空泡样改变;与假手术组比较,模型组和活血通督组NGF和GFAP表达均明显升高(P<0.01);与模型组比较,活血通督组NGF阳性细胞数表达较高(P<0.01),GFAP阳性表达较低(P<0.01)。结论:活血通督汤能促进大鼠急性脊髓损伤后NGF的表达,抑制GFAP的表达,从而有利于急性脊髓损伤的修复。
Objective: To observe the effect of Huoxuetongtang on expression of nerve growth factor (NGF) and glial fibrillary acidic protein (GFAP) after acute spinal cord injury and to explore the mechanism of action of Huoxuetong decoction on spinal cord injury. Methods: Twenty-four adult female Sprague-Dawley rats were randomly divided into sham operation group, model group and Huoxutongsuotang group, with 8 rats in each group. Preoperative and postoperative 7d were given continuous gavage, Huoxuetongtu group was fed Huoxuetongsuitang, the other two groups were fed 0.9% sodium chloride solution, both for 2 times a day. The sham-operated group was exposed to the spinal cord, but the spinal cord was not impacted. The model group and the Huoxuetongdu decoction group were given modified Allen’s method to prepare the spinal cord injury model. 7d after surgery, perfusion fixed, embedded in paraffin. The morphological changes of spinal cord and neurons were observed by Nissl staining. The expression of NGF and GFAP in spinal cord was detected by immunofluorescence. Results: The gray matter in the sham operation group showed a butterfly shape with a complete structure, a clear demarcation of the gray matter, and a plaque-like Nissl body in the cytoplasm. Compared with the model group, the blood stasis area was smaller and the neuron edema was lighter, Compared with sham group, the expression of NGF and GFAP in model group and Huoxuetongdu group were significantly increased (P <0.01); Compared with model group, the expression of NGF The number of positive cells was higher (P <0.01), while the positive expression of GFAP was lower (P <0.01). Conclusion: Huoxue Tongdu Tang can promote the expression of NGF and inhibit the expression of GFAP after acute spinal cord injury in rats, which is good for the repair of acute spinal cord injury.
