目的:观察兔冷冻卵巢组织子宫系膜、卵巢囊和卵巢内多点种植后组织形态及功能,探讨移植部位和移植方法。方法:15只新西兰白兔随机分为3组,每组5只:1组,新鲜卵巢组织子宫系膜及卵巢囊种植;2组:冷冻卵巢组织子宫系膜及卵巢囊种植;3组:冷冻卵巢组织卵巢内种植。通过卵巢组织光镜和电镜观察、子宫内膜组织光镜观察、阴道脱落细胞学检查,反映种植后卵巢组织存活、卵泡生长发育及内分泌功能等情况。结果:冷冻复苏组织和新鲜组织相比,2组、3组移植后组织与冷冻复苏组织相比,3个移植组间相比,正常卵泡和形态改变卵泡所占比率差异均无统计学意义(P>0.05);3个移植组的正常卵泡比率均低于新鲜组织(P<0.05);3个移植组卵巢组织中存在正常比例的各级卵泡,并且近成熟卵泡的比率与新鲜卵巢组织相比差异无统计学意义(P>0.05),2组、3组移植后组织与冷冻复苏组织比较,近成熟卵泡所占比率增加(P<0.05);冷冻后和移植后卵巢组织形态和超微结构无明显改变。结论:小块卵巢组织冷冻可以获得良好的冷冻保存效果;冷冻卵巢组织自体多点种植后,卵巢组织光镜形态学和超微结构无明显改变,卵泡存活并正常发育;卵巢组织种植于子宫系膜、卵巢囊和卵巢内均可以得到良好的效果。
OBJECTIVE: To observe the morphology and function of multi-point mesentery, ovary and ovary in frozen ovary tissue of rabbits, and to investigate the transplanted sites and transplantation methods. Methods: Fifteen New Zealand white rabbits were randomly divided into three groups with 5 rats in each group: group 1, fresh ovarian tissue, uterine mesangial and ovarian cysts; group 2: frozen uterine mesangial and ovarian cysts; group 3: Ovarian tissue in the ovary. Observed by light microscope and electron microscope in ovary tissue, endometrial tissue by light microscopy, vaginal cytology examination to reflect the survival of ovarian tissue after planting, follicular growth and development and endocrine function. Results: Compared with frozen-thawed tissue, the rates of normal follicles and morphologically-modified follicles in frozen-thawed tissue were not significantly different from those in fresh tissue (P <0.05). The normal follicles in three transplantation groups were lower than that in fresh tissue (P <0.05). The normal ovarian follicles in three transplantation groups were normal, and the ratio of nearly mature follicles and fresh ovarian tissue (P> 0.05). Compared with the frozen-thawed tissue, the ratio of mature follicles in the 2 groups and 3 groups increased (P <0.05). The morphology and ultrastructure of the ovary after freezing and transplantation Structure did not change significantly. Conclusion: The cryopreservation of small ovarian tissue can be well preserved. After the multi-point implantation of frozen ovarian tissue, the morphological and ultrastructural changes of the light microscope in ovary tissue have no obvious changes, the ovarian tissue survived and developed normally. The ovarian tissue was planted in the uterus Membrane, ovarian cysts and ovaries can get good results.
