目的:观察补阳还五汤对单侧输尿管结扎肾间质纤维化大鼠模型α-平滑肌肌动蛋白(α-SMA)的作用。方法:将SD大鼠随机分为单侧输尿管结扎造模组(UUO)和假手术对照组。造模大鼠再随机分为模型组、补阳还五汤治疗组(中药组)和依那普利组。大鼠分别在第7,14,28天进行肾功能、HE染色、Masson染色和免疫组化检测。结果:与假手术组相比,模型组大鼠肾功能显著下降;依那普利和中药干预后,肾功能显著提高。光镜显示模型组大鼠出现显著间质纤维化,依那普利和中药干预显著减轻了肾小管损害。各个时间点,模型组肾间质损伤指数和α-SMA的表达与假手术组相比,显著升高。通过依那普利和中药干预在第7天时,显著降低肾间质损伤指数(模型组3.88±0.15,依那普利组2.48±0.43,中药组2.54±0.37,P<0.05)和α-SMA的表达(模型组7.27±1.62,依那普利组5.27±2.18,中药组5.05±1.45,P<0.05);用药第14,28天后,可进一步降低肾间质损伤指数(14 d,模型组7.65±0.87,依那普利组4.17±0.39,中药组4.24±0.42,P<0.01;28 d,模型组10.86±1.49,依那普利组6.19±0.58,中药组6.94±0.77,P<0.01)和α-SMA的表达(14 d,模型组17.48±4.16,依那普利组10.15±2.85,中药组10.58±3.19,P<0.01;28 d,模型组29.68±7.24,依那普利组17.81±0.58,中药组18.04±4.72,P<0.01)。结论:补阳还五汤可改善单侧输尿管结扎大鼠肾功能和肾小管间质纤维化,降低肾间质损伤指数和α-SMA的表达,对拮抗肾间质纤维化有一定的作用。
OBJECTIVE: To observe the effect of Buyang Huanwu decoction on α-smooth muscle actin (α-SMA) in rats with unilateral ureteral ligation of renal interstitial fibrosis. Methods: SD rats were randomly divided into unilateral ureteral ligation model group (UUO) and sham-operated control group. Model rats were randomly divided into model group, Buyang Huanwu Decoction group (TCM group) and enalapril group. Kidney function, HE staining, Masson staining and immunohistochemistry were performed on the 7th, 14th and 28th day respectively. RESULTS: Compared with the sham-operated group, the renal function of the model group was significantly decreased. After intervention with enalapril and Chinese medicine, the renal function was significantly increased. Light microscopy showed that the rats in the model group had significant interstitial fibrosis, and intervention with enalapril and Chinese herbs significantly reduced renal tubular damage. At each time point, the renal interstitial injury index and the expression of α-SMA in the model group were significantly higher than those in the sham group. The renal interstitial injury index (model group 3.88±0.15, enalapril group 2.48±0.43, Chinese medicine group 2.54±0.37, P<0.05) and α-SMA were significantly decreased by enalapril and Chinese medicine intervention on the 7th day. Expression (model group, 7.27±1.62, enalapril group, 5.27±2.18, Chinese medicine group, 5.05±1.45, P<0.05); on the 14th and 28th days after administration, renal interstitial injury index could be further reduced (14 d, model group 7.65 ±0.87, 4.17 ± 0.39 in enalapril group, 4.24 ± 0.42 in Chinese herb group, P <0.01; 28 d, 10.86 ± 1.49 in model group, 6.19 ± 0.58 in enalapril group, and 6.94 ± 0.77 in Chinese herb group, P <0.01) And α-SMA expression (14 d, model group, 17.48±4.16, enalapril group, 10.15±2.85, Chinese medicine group, 10.58±3.19, P<0.01; 28 d, model group, 29.68±7.24, enalapril group, 17.81 ±0.58, Chinese medicine group 18.04±4.72, P<0.01). Conclusion: Buyang Huanwu Decoction can improve the renal function and tubulointerstitial fibrosis in rats with unilateral ureteral ligation, reduce the renal interstitial injury index and the expression of α-SMA, and has a certain role in antagonizing renal interstitial fibrosis.
