本文介绍用化学修饰的方法研究植物病毒(Kcnnedya黄花叶病毒)核糖核酸3′-末端的核苷酸顺序。应用(α-~(32)p)-ATP标记后以聚丙烯酰胺凝胶电泳制备和纯化所需要的核苷酸片段,它们系来自于RNA分子3′-末端大约100~200个核苷酸残基长度的片段。根据Peattie直接化学定序法测定它的核苷酸顺序。已经确定Kennedya黄花叶病毒核糖核酸3′-末端区域的84个核苷酸的顺序并根据这些实验结果提示它的二级结构模型。这个模型的主要特点是与典型的tRNA分子三叶草结构不问,但与芜菁黄花叶病毒核糖核酸3′-末端的tRNA样结构相似。
This article describes the chemical modification of the nucleotide sequence of the 3’-end of the plant virus (Kcnnedya yellow mosaic virus) RNA. The desired nucleotide fragments were prepared and purified by polyacrylamide gel electrophoresis after labeling with (α ~ (32) p) -ATP from about 100 to 200 nucleotides at the 3’-end of the RNA molecule Residue length fragments. Its nucleotide sequence was determined by Peattie direct chemical sequencing. The 84 nucleotide sequence of the 3’-terminal region of Kennedy’s yellow mosaic virus RNA has been determined and suggests its secondary structure model based on these experimental results. The main feature of this model is that it is not related to the typical tRNA molecule clover structure, but similar to the tRNA-like structure at the 3’-end of the turnip yellow mosaic virus RNA.
