目的:探讨胰岛素作用下,牙周膜(PDL)细胞碱性磷酸酶(ALP)活性和总蛋白含量的变化。方法:细胞生物学法、酶动力学法和考马斯亮蓝法。结果:胰岛素浓度超过10-4U/ml时,明显增加PDL细胞的ALP活性和总蛋白含量(P<0.01)。其中,在10-4~10-1U/ml之间,效应随浓度增加而增加,超过10-1U/ml效应趋于稳定,此时效应最大,ALP活性和总蛋白含量分别比对照组增加1.4倍和1.2倍。结论:胰岛素明显促进PDL细胞分化及蛋白合成功能。胰岛素作为骨代谢的重要调节因子,可能对牙周组织的再生修复有调节作用。
Objective: To investigate the changes of alkaline phosphatase (ALP) activity and total protein in periodontal ligament (PDL) cells under the action of insulin. Methods: Cell biology, enzyme kinetics and Coomassie brilliant blue method. Results: When insulin concentration exceeded 10-4U / ml, the ALP activity and total protein content of PDL cells were significantly increased (P <0.01). Among 10-4 U / ml, the effect increased with the increase of concentration, while the effect of more than 10-1 U / ml tended to be stable, the effect was maximal, ALP activity and total protein content increased by 1 .4 times and 1.2 times. Conclusion: Insulin can significantly promote PDL cell differentiation and protein synthesis. Insulin as an important regulator of bone metabolism may have regulative effects on the regeneration of periodontal tissue.
