目的:探讨依那普利和丹芪合剂对糖尿病大鼠肾组织结缔组织生长因子(CTGF)及蛋白激酶Cα(PKCα)表达的影响。方法:SD大鼠随机分为正常组(A组)、糖尿病组(B组)、糖尿病丹芪合剂组(C组)和依那普利组(D组)。尾静脉注射链脲佐菌素复制糖尿病模型。用免疫组化检测肾组织CTGF、PKCα、纤维连接蛋白(FN);Western印迹法检测肾组织CTGF蛋白水平;光镜观察肾组织的形态结构变化;生化方法检测尿蛋白、血糖、血脂和血肌酐。结果:12周后,药物处理的糖尿病大鼠肾组织CTGF、PKCα、FN表达高于正常组,但低于未处理的糖尿病组,血肌酐和尿蛋白水平较低,肾脏病理改变较轻,同时CTGF与PKCα和FN及肾脏指数和肾功能呈显著正相关。丹芪合剂组动物与未处理组相比,血脂水平较低。结论:依那普利和丹芪合剂可能通过PKC信号通路而抑制CTGF的表达,使FN的生成减少,从而对糖尿病大鼠肾脏起保护作用。
Objective: To investigate the effects of enalapril and Danqi mixture on the expression of connective tissue growth factor (CTGF) and protein kinase Cα (PKCα) in diabetic rats. Methods: SD rats were randomly divided into normal group (group A), diabetic group (group B), diabetic Danqi mixture group (group C) and enalapril group (group D). Tail vein injection of streptozotocin in the replication of diabetic model. Immunohistochemistry was used to detect the expression of CTGF, PKCα and fibronectin (FN) in renal tissue; Western blotting was used to detect the level of CTGF protein in kidney; morphological changes of renal tissue were observed by light microscopy; urinary protein, blood glucose, serum lipid and serum creatinine . Results: After 12 weeks, the expression of CTGF, PKCα and FN in renal tissue of drug-treated diabetic rats was higher than that of normal rats, but lower than that of untreated diabetic rats. The levels of serum creatinine and urinary protein were lower and pathological changes of kidney were lighter. There was a significant positive correlation between CTGF, PKCα and FN, kidney index and renal function. Danqi mixture group of animals compared with untreated group, low blood lipid levels. Conclusion: Enalapril and Danqi Mixture can inhibit the expression of CTGF through the PKC signaling pathway and decrease the production of FN, thus protecting the kidneys of diabetic rats.
