目的:观察龙葵鲜药对人鼻咽癌细胞(CNE-1)增殖和凋亡/坏死的影响。方法:制备龙葵鲜药水煎液1g/ml,体外培养CNE-1细胞,并取对数生长期细胞分别加入浓度为5、10、30、50、80mg/ml的龙葵鲜药水煎液,即为不同浓度的龙葵鲜药水煎液干预组,对照组不加药;采用MTS法分别观察龙葵鲜药水煎液作用CNE-1细胞24h、48h和72h后的抑制率,并采用流式细胞术检测各组药物对CNE-1细胞凋亡/坏死的影响。结果:5、10、30、50、80mg/ml龙葵鲜药水煎液对CNE-1细胞增殖均呈现明显的抑制作用,而且随着给药浓度的增加,细胞的凋亡和坏死明显升高。结论:本研究证实了龙葵鲜药水煎液在体外有效抑制CNE-1细胞的增殖和诱导CNE-1的凋亡和坏死,从而发挥其抗肿瘤作用。
Objective: To observe the effect of Solanum nigrum on proliferation and apoptosis / necrosis of human nasopharyngeal carcinoma (CNE-1) cells. Methods: Prepared Solanum nigrum decoction 1g / ml, cultured in vitro CNE-1 cells, and logarithmic growth phase were added to the concentration of 5,10,30,50,80 mg / ml of Solanum nigrum decoction, The effect of different concentrations of Solanum nigrum decoction intervention group, the control group without drug; MTS method were used to observe the effect of Solanum nigrum freshwater decoction of CNE-1 cells 24h, 48h and 72h after inhibition, and the use of flow Cytometry was used to detect the effect of each drug on the apoptosis / necrosis of CNE-1 cells. Results: The decoctions of 5, 10, 30, 50, 80 mg / ml Solanum nigrum could significantly inhibit the proliferation of CNE-1 cells, and the apoptosis and necrosis of CNE-1 cells were significantly increased with the increase of drug concentration . Conclusion: This study confirmed that Solanum nigrum decoction can effectively inhibit CNE-1 cell proliferation and induce apoptosis and necrosis of CNE-1 cells in vitro and thus exert its anti-tumor effect.
