目的:定量分析膨大弯颈霉菌株SFYT002菌丝体中核苷类成分。方法:本研究从新疆细虫草菌核分离出一株膨大弯颈霉菌株SFYT002,以已知的7种核苷为标准品,并与野生新疆细虫草和冬虫夏草做比较,采用高效液相色谱法(HPLC)对其核苷类成分进行测定。结果:膨大弯颈霉SFYT002菌株含有与冬虫夏草相同的7种核苷类成分,其中该菌株尿嘧啶为75.668μg·g-1,略高于冬虫夏草,为新疆细虫草的4倍左右;肌苷含量达到663.776μg·g-1,为冬虫夏草3倍,为新疆细虫草的27倍之多;腺苷含量114.653μg·g-1,分别为冬虫夏草和新疆细虫草腺苷含量的1/5和2/5左右;尿苷、鸟苷、腺嘌呤含量也接近冬虫夏草,高于新疆细虫草。此外,在膨大弯颈霉SFYT002菌株中检测出虫草素,含量为32.261μg·g-1,但新疆细虫草和冬虫夏草中未检测出虫草素,为虫草素开发提供了一种新的药源。结论:膨大弯颈霉菌株SFYT002核苷类成分含量丰富,具有开发潜力和前景。
Objective: To quantitatively analyze nucleosides in mycelium of SFYT002. Methods: In this study, a strain of Penicillium funicolosus SFYT002 was isolated from Xinjiang Cordyceps sinensis. Seven known nucleosides were used as standards and compared with wild Xinjiang Cordyceps sinensis and Cordyceps sinensis. High performance liquid chromatography (HPLC) to determine its nucleoside component. Results: The strains of SFYT002 had the same seven nucleosides as Cordyceps Sinensis, of which the concentration of uracil was 75.668μg · g-1, which was slightly higher than that of Cordyceps Sinensis and about 4 times that of Cordyceps sinensis. The content of inosine Reached 663.776μg · g-1, which was 3 times of that of Cordyceps sinensis and 27 times of that of Xinjiang Cordyceps; the content of adenosine was 114.653μg · g-1, which were respectively 1/5 and 2 / 5; uridine, guanosine, adenine content is also close to Cordyceps sinensis, Xinjiang Cordyceps than higher. In addition, cordycepin was detected in the SFYT002 strain with the content of 32.261μg · g-1, but cordycepin was not detected in Xinjiang Cordyceps sinensis and Cordyceps sinensis, which provided a new source of drug for the development of cordycepin. CONCLUSION: The strain nucleocapsid of SFYT002 is characterized by its richness of development and potential.
