[目的]研究Y染色体微卫星基因座DYS441,DYS442在吉林省朝鲜族和汉族男性人群中的分布特征.[方法]采用PCR扩增结合聚丙烯酰胺凝胶电泳技术和银染显带方法对205名朝鲜族男性和207名汉族男性DYS441,DYS442基因座进行检测,并用Arlequin 2000软件进行数据分析.[结果]在朝鲜族男性人群DYS441基因座中检测到8个等位基因,个人识别能力为0.749 3,DYS442基因座中检测到7种等位基因,个人识别能力为0.508 3;在汉族男性人群DYS441基因座中检测到8个等位基因,个人识别能力为0.803 1,DYS442基因座中检测到5种等位基因,个人识别能力为0.627 3.分析由2个基因座共同构成的单倍型,在朝鲜族人群中检测到23种,在汉族人群中为31种,其单倍型变异度分别为0.874 6和0.927 2.[结论]DYS441和DYS442基因座具有较高的遗传多态性,是较理想的遗传标志,可为朝鲜族和汉族人群法医学个人识别、亲权鉴定及父系遗传学研究提供基础资料.
[Objective] The research aimed to investigate the distribution characteristics of Y chromosome microsatellite loci DYS441 and DYS442 in Korean and Han male population in Jilin Province. [Methods] PCR amplification combined with polyacrylamide gel electrophoresis and silver staining And Korean male DYS441 and DYS442 loci were detected and analyzed by Arlequin 2000 software. [Results] Eight alleles were detected in DYS441 locus of Korean male population, and their individual recognition ability was 0.749 3, 7 alleles were detected in DYS442 loci with an individual recognition ability of 0.508 3; 8 alleles were detected in DYS441 locus in Han male population, the individual recognition ability was 0.803 1, DYS442 locus detected Five alleles were identified with an individual identification ability of 0.627. 3. The haplotypes consisted of two loci were analyzed. Twenty-three species were detected in the Korean population and 31 species in the Han population. The haplotype variation Respectively 0.874 6 and 0.927 2. [Conclusion] The DYS441 and DYS442 loci have high genetic polymorphisms and are ideal genetic markers for forensic identification, paternity testing and paternal inheritance in Korean and Han populations Study provides basic data.
