目的研究H.pylori作用后肝细胞系HepG2蛋白质表达谱的改变,通过对差异蛋白质点的鉴定与分析,以探讨H.pylori对肝细胞的病理作用。方法采用体外肝细胞和H.pylori共培养,应用双向凝胶电泳分离H.pylori处理前后细胞总蛋白,筛选出差异表达的蛋白质点,并进行质谱分析鉴定。结果鉴定出7种表达上调的蛋白质点,包括整合素-β1、蛋白激酶C-α、LIM同源盒蛋白Lhx1、真核翻译起始因子2-β亚单位、PINCH蛋白、MAPK激酶3、Ras相关蛋白Rab-37等。这些蛋白质参与了基因表达调控、细胞免疫、细胞生长、信号传导等众多事件。结论H.pylori可能通过介导这些蛋白质的上调而发挥对HepG2细胞的病理作用。这种蛋白质组的差异分析有助于进一步研究H.pylori与人类肝胆疾病的关系。
Objective To study the change of HepG2 protein expression in hepatocellular carcinoma cell line after treated with H.pylori, and to identify and analyze the differential protein spots to explore the pathological effect of H.pylori on hepatocytes. Methods Co-culture of hepatocytes and H.pylori in vitro was performed. Two-dimensional gel electrophoresis was used to separate the total proteins of the cells before and after H.pylori treatment. The differentially expressed protein spots were screened out and identified by mass spectrometry. RESULTS: Seven of the up-regulated protein spots were identified including integrin-β1, protein kinase C-α, LIM homeobox protein Lhx1, eukaryotic translation initiation factor 2-β subunit, PINCH protein, MAPK kinase 3, Related protein Rab-37 and so on. These proteins are involved in numerous events such as gene expression regulation, cellular immunity, cell growth, signaling and so on. Conclusion H.pylori may play a pathological role in HepG2 cells by mediating the upregulation of these proteins. The differential analysis of this proteome will help further study the relationship between H.pylori and human hepatobiliary diseases.
