目的 探讨血管内皮生长因子 (VEGF)在结肠癌中的表达及其与微血管密度 (MVD)和p5 3之间的关系。方法 用免疫组化SABC法检测 6 8例结肠癌组织不同区域VEGF、MVD和 p5 3的阳性表达。结果 结肠癌区VEGF、MVD和 p5 3的表达明显高于癌旁区和正常区。结肠癌组织VEGF及p5 3的表达与肿瘤浸润深度、淋巴结转移、远处转移、血管侵犯及Dukes分期密切相关 ,而与组织学分型无关。p5 3(+ )或VEGF(+ )组MVD(34.6± 12 .2 ;31.2± 12 .6 )均显著高于p5 3(- )或VEGF(- )组(15 .0± 7.9;12 .7± 6 .3,P <0 .0 1) ;VEGF和 p5 3均为阳性时 ,MVD值最大 (36 .5± 11.9,P <0 .0 1)。MVD记数与VEGF表达明显相关 (P <0 .0 1) ,p5 3表达与VEGF表达和MVD记数均显著相关 (P <0 .0 1)。结论 在结肠癌血管生成过程中 ,可能存在 p5 3 VEGF调节旁路 ,p5 3基因在调控肿瘤血管形成方面起重要作用。
Objective To investigate the expression of vascular endothelial growth factor (VEGF) in colon cancer and its relationship with microvessel density (MVD) and p53. Methods Immunohistochemical SABC method was used to detect the positive expression of VEGF, MVD and p53 in different tissues of 68 cases of colon cancer. Results The expression of VEGF, MVD and p53 in the colon cancer was significantly higher than that in the paracancerous and normal areas. The expression of VEGF and p53 in colon cancer tissues was closely related to the depth of tumor invasion, lymph node metastasis, distant metastasis, vascular invasion, and Dukes stage, but not histological type. The MVD in the p53(+) or VEGF(+) group was significantly higher than that of the p53(-) or VEGF(-) group (35.0±12.2; 31.2±12.6) (15.0±7.9; 12.7%). ± 6.3, P <0.01); VEGF and p53 were positive, MVD was the highest (36. 5 ± 11.9, P <0.01). MVD scores were significantly correlated with VEGF expression (P < 0.01), and p53 expression was significantly correlated with VEGF expression and MVD count (P <0.01). Conclusion In the process of angiogenesis of colon cancer, p53 VEGF regulation bypass may exist, and p53 gene plays an important role in the regulation of tumor angiogenesis.
