精氨酸酶,又名L-精氨酸尿素水解酶,是一种将精氨酸水解成鸟氨酸和尿素的水解酶,本身为内源性蛋白酶,在人体内尿素循环中起到重要作用。工业上可利用其水解活性以精氨酸生产鸟氨酸。另外,精氨酸酶还有着显著的抑制肿瘤细胞分裂的作用[1~2]。但是,在临床应用中,存在以下缺点:体内循环半衰期短,易被循环系统清除,需要多次给药等问题,给患者的日常生活带来很大的不便。而聚乙二醇(Polyethylene Glycol,PEG)化学修饰能够降低蛋白免疫原性以及有效延长药物半衰期,是解决上述问题的有效途径之一[3]。
Arginase, also known as L-arginine urea hydrolase, is a hydrolase that hydrolyzes arginine into ornithine and urea, itself an endogenous protease that plays an important role in urea circulation in the human body effect. Arginine can be used to produce ornithine industrially for its hydrolytic activity. In addition, arginase also has a significant role in inhibiting tumor cell division [1 ~ 2]. However, there are some shortcomings in clinical application such as short half-life of systemic circulation, easy removal by the circulatory system and multiple administrations, which bring great inconvenience to patients’ daily life. However, the chemical modification of polyethylene glycol (PEG) can reduce the immunogenicity of protein and effectively prolong the half-life of drug, which is an effective way to solve the above problems [3].
