目的观察脂肪干细胞对顺铂所致卵巢早衰大鼠细胞因子表达的影响,探讨大鼠脂肪干细胞对卵巢早衰的治疗作用。方法选取8~10周龄的SD大鼠,采用连续7 d腹腔注射顺铂,制备卵巢早衰大鼠模型。实验设计为顺铂造模卵巢早衰组、大鼠脂肪干细胞治疗组和空白对照组。大鼠脂肪干细胞治疗组在造模后连续腹腔注射大鼠脂肪干细胞7 d,治疗后采用化学发光法检测大鼠血清雌二醇(E_2)及卵泡刺激素(FSH)水平;应用免疫组织化学法检测GnRHR蛋白表达。结果卵巢早衰大鼠经过脂肪干细胞治疗后,大鼠血清E_2及FSH分泌趋于恢复(P<0.05);与顺铂造模卵巢早衰组相比,大鼠脂肪干细胞治疗组能明显上调颗粒细胞GnRHR蛋白表达(P<0.05)。结论大鼠脂肪干细胞能减轻顺铂对卵巢的损伤,其机制可能是通过促进各级卵泡发育,降低FSH水平,升高E_2水平,调节颗粒细胞GnRHR蛋白表达,从而改善卵巢功能,减轻顺铂对卵巢的损伤,发挥治疗卵巢早衰的作用。
Objective To observe the effect of adipose-derived stem cells on the expression of cytokines in rats with premature ovarian failure induced by cisplatin and to explore the therapeutic effect of adipose-derived stem cells on premature ovarian failure in rats. Methods Sprague-Dawley (SD) rats aged 8 to 10 weeks were selected and treated with cisplatin (7 days) intraperitoneally. Experimental design for cisplatin model of premature ovarian failure group, rat adipose-derived stem cell treatment group and blank control group. Rat adipose-derived stem cells were injected intraperitoneally with rat’s adipose-derived stem cells for 7 days. The levels of serum estradiol (E2) and follicle-stimulating hormone (FSH) were measured by chemiluminescence assay. The expression of FSH was detected by immunohistochemistry GnRHR protein expression was detected. Results Secretion of E_2 and FSH in serum of rats with premature ovarian failure after ADSCs treatment tended to recover (P <0.05). Compared with Cisplatin-induced premature ovarian failure group, Adipose-derived stem cells treatment group of rats could significantly upregulate GnRHR Protein expression (P <0.05). Conclusion Adipose-derived stem cells can attenuate ovarian damage induced by cisplatin in rats. The mechanism may be through promoting follicular development at all levels, decreasing FSH level, increasing E_2 level and regulating GnRHR protein expression in granulosa cells, thereby improving ovarian function and reducing cisplatin-induced Ovarian damage, play a role in the treatment of premature ovarian failure.
