目的探讨福辛普利对肾间质纤维化的防治作用。方法采用单侧输尿管结扎法致肾间质纤维化的动物模型,以福辛普利为阳性对照,测血肌酐(Scr)、尿素氮(BUN)、血浆血管紧张素Ⅱ(AngⅡ),免疫组化和原位杂交方法分别检测肾组织中基质金属蛋白酶组织抑制剂-1(TIMP-1)、纤维连接蛋白(FN)和TIMP-1mRNA的表达。结果福辛普利能降低血Scr、BUN、AngⅡ水平,显著抑制TIMP-1表达上调,减轻肾间质纤维化程度。结论福辛普利有早期防治肾间质纤维化作用。
Objective To investigate the preventive and therapeutic effects of fosinopril on renal interstitial fibrosis. Methods Animal models of renal interstitial fibrosis induced by unilateral ureteral ligation were established. Fosinopril was used as a positive control, and serum creatinine (Scr), blood urea nitrogen (BUN), plasma angiotensin Ⅱ (AngⅡ) (TIMP-1), fibronectin (FN) and TIMP-1 mRNA in renal tissue were detected by in situ hybridization and in situ hybridization. Results Fosinopril could decrease the levels of Scr, BUN and AngⅡ in blood, significantly up-regulate the expression of TIMP-1 and decrease the degree of renal interstitial fibrosis. Conclusion Fosinopril has early prevention and treatment of renal interstitial fibrosis.
