目的分析高猪油日粮及高蔗糖日粮分别诱发胰岛素抵抗大鼠的血液生化指标差异,为此类模型的建立及实验研究提供参考。方法雄性SD大鼠随机分为3组,对照组给予普通日粮,高脂组给予高猪油含量的日粮,高糖组给予高蔗糖含量的日粮。喂养6周,每两周测定空腹血糖、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-c)、总胆固醇(TCH)、胰岛素,根据胰岛素敏感性指数(ISI)=ln1/(FPG×FINS)评定大鼠的胰岛素敏感性。结果 6周后,高脂、高糖组ISI显著低于对照组(P<0.05);猪油组血清胰岛素显著高于对照组(P<0.05)、血清HDL-c显著低于对照组(P<0.05);高蔗糖组血糖、胰岛素均上升,但与对照组无统计学差异;而各组间血清总胆固醇、甘油三酯及体重无明显差异(P>0.05)。结论高脂、高糖日粮均可诱发IR大鼠模型,高脂模型具有血清胰岛素显著升高、HDL-c显著降低的特点而高糖模型伴有较高的血糖、胰岛素。
OBJECTIVE: To analyze the difference of blood biochemical indexes in insulin resistance rats induced by high lard oil diet and high sucrose diet, and provide references for the establishment of such models and experimental study. Methods Male Sprague-Dawley rats were randomly divided into three groups. The control group was given normal diet, the high-fat group was given the diet with high lard oil content, and the high-sugar group was given the diet with high sucrose content. Fasting blood glucose, triglyceride (TG), high density lipoprotein cholesterol (HDL-c), total cholesterol (TCH), insulin and insulin sensitivity according to Insulin Sensitivity Index (ISI) = ln1 / (FPG × FINS) to assess insulin sensitivity in rats. Results After 6 weeks, the ISI in high-fat and high-sugar group was significantly lower than that in control group (P <0.05); the serum insulin in lard oil group was significantly higher than that in control group (P <0.05), and the serum HDL-c was significantly lower than that in control group <0.05). There was no significant difference between the high sucrose group and the control group, but there was no significant difference in serum total cholesterol, triglyceride and body weight between the two groups (P> 0.05). Conclusion The model of IR rats can be induced by high-fat and high-sugar diet. High-fat model has markedly elevated serum insulin and HDL-c, while high-glucose model has higher blood sugar and insulin.
