研究了尖顶羊肚菌菌种在不同条件下的短期保藏对其菌丝生长的影响,以寻求适宜的菌种保藏条件。采用10℃/5℃(昼/夜)和15℃/10℃2种变温条件、散射光和黑暗2种光照条件,分别保藏菌种30 d和60 d,以冰箱(4℃)保藏为对照。结果表明,10℃/5℃保藏时,尖顶羊肚菌的菌丝生长优于15℃/10℃及对照保藏;散射光保藏菌落颜色和菌丝气生性好于黑暗保藏;在10℃/5℃温度下,保藏时间对菌丝生长影响不大,可以继续延长保藏时间,在15℃/10℃温度下,不宜超过60 d的保藏时间。因此,较适宜的保藏条件是10℃/5℃、散射光、保藏30 d,菌株表现为菌落边缘整齐,菌丝气生性中等,菌丝致密,菌落角变面积小,菌丝长速均匀稳定和菌丝干重较高。说明尖顶羊肚菌菌种在较低的常温、有散射光条件下保藏比保藏在冰箱里效果佳。
The effects of short-term preservation of Morchella esculenta on mycelial growth under different conditions were studied in order to find suitable storage conditions for the strain. Two kinds of illumination conditions of 10 ℃ / 5 ℃ (day / night) and 15 ℃ / 10 ℃ were used, and the light and dark conditions were scattered for 30 days and 60 days respectively. The samples were stored in refrigerator (4 ℃) . The results showed that the mycelial growth of Morchella esculentus was better than that of the control at 15 ℃ / 10 ℃ when stored at 10 ℃ / 5 ℃. The colony color and mycelia of the colony were better than those of the dark storage at 10 ℃ / 5 ℃, the storage time has little effect on the mycelium growth, and can prolong the storage time. At the temperature of 15 ℃ / 10 ℃, the storage time should not exceed 60 days. Therefore, the most suitable storage conditions were light scattering at 10 ℃ / 5 ℃ for 30 days. Strains of the colonies showed neat edges, moderate mycelial aeration, dense mycelium, small area of the colonies and uniform long-term hyphae And hyphae dry weight higher.
