目的:探讨CD25基因多态性与云南汉族人群肺癌发生发展的相关性。方法:选取云南地区汉族人群肺癌Ⅰ+Ⅱ期患者151例,肺癌Ⅲ+Ⅳ期患者316例,健康体检人群384例,采用Taq Man探针基因分型方法对CD25基因中2个多态性位点rs7072793(C>T)和rs3118470(C>T)进行基因分型,研究其等位基因、基因型及所构建的单倍型在肺癌Ⅰ+Ⅱ期患者、肺癌Ⅲ+Ⅳ期患者和健康对照人群中频率分布的差异。结果:rs7072793和rs3118470位点等位基因在各组中的分布频率比较,差异无统计学意义(P>0.05),两个位点等位基因可能与肺癌的发生发展无关;遗传模式分析的结果经年龄校正后,rs7072793位点采用最优遗传模式Recessive分析发现,基因型CC-CT和TT在Ⅰ+Ⅱ期组和Ⅲ+Ⅳ期组中的分布频率比较,差异有统计学意义(P=0.004);rs3118470位点采用最优遗传模式Dominant分析发现,基因型CT-CC和TT基因型在Ⅰ+Ⅱ期组和Ⅲ+Ⅳ期组中分布频率比较,差异有统计学意义(P=0.019);rs7072793位点基因型CC-CT可能是肺癌发生的保护性因素(OR=0.45,95%CI:0.25~0.80),rs3118470位点基因型TT可能是肺癌发生的风险因素(OR=1.79,95%CI:1.09~2.95)。结论:CD25基因中多态性位点rs7072793位点基因型CC-CT可能是肺癌发生的保护性因素,rs3118470位点基因型TT可能是肺癌发生的风险因素。
Objective: To investigate the association between CD25 gene polymorphism and the occurrence and development of lung cancer in Yunnan Han population. Methods: One hundred and fifteen patients with stage Ⅰ + Ⅱ lung cancer in Han nationality in Yunnan province, 316 patients with stage Ⅲ + Ⅳ lung cancer and 384 healthy subjects were enrolled in this study. Taq Man probe genotyping was used to detect the polymorphisms of two polymorphic sites The genotypes of rs7072793 (C> T) and rs3118470 (C> T) were genotyped. The alleles, genotypes and haplotypes were determined in patients with stage Ⅰ + Ⅱ lung cancer, stage Ⅲ + Ⅳ lung cancer and health Differences in frequency distribution in control population. Results: There was no significant difference in the distribution frequency of rs7072793 and rs3118470 among the groups (P> 0.05). The alleles of the two loci were probably not related to the occurrence and development of lung cancer. The results of genetic model analysis After age correction, rs7072793 locus using the optimal genetic model Recessive analysis found that genotypes CC-CT and TT in the Ⅰ + Ⅱ group and Ⅲ + Ⅳ group frequency distribution, the difference was statistically significant (P = 0.004). The distribution of genotype CT-CC and TT genotypes in genotypes Ⅰ + Ⅱ and Ⅲ + Ⅳ was significantly higher than rs3118470 (P = 0.019) (OR = 0.45, 95% CI: 0.25-0.80). Genotype TT of rs3118470 locus may be a risk factor for lung cancer (OR = 1.79, 95% CI: 1.09 ~ 2.95). Conclusion: The genotype CC-CT at rs7072793 site of CD25 gene may be a protective factor in lung cancer. Genotype TT of rs3118470 may be a risk factor for lung cancer.
