目的探讨103钯(103Pd)释放的γ射线治疗胆管癌的作用机制。方法用MTT法分析103Pd释放的γ射线对胆管癌细胞生长的影响,用电镜、DNA凝胶电泳和流式细胞仪观察细胞形态和凋亡率的变化。结果103Pd释放的γ射线明显抑制胆管癌细胞生长。5.333 mci剂量的γ射线诱导出典型的细胞凋亡特征,如凋亡小体、特征性亚G1峰等。6.645 mci以上剂量的γ射线导致细胞坏死。结论γ射线诱导胆管癌细胞凋亡可能是放射治疗胆管癌的重要机制之一,103Pd释放的γ射线诱导胆管癌细胞凋亡与照射剂量有密切关系。
Objective To investigate the mechanism of γ-rays released by 103Pd (103Pd) in the treatment of cholangiocarcinoma. Methods MTT assay was used to analyze the effects of γ-rays released by 103Pd on the growth of cholangiocarcinoma cells. The changes of cell morphology and apoptosis rate were observed by electron microscopy, DNA gel electrophoresis and flow cytometry. Results 103Pd release of γ-rays significantly inhibited the growth of cholangiocarcinoma cells. 5.333 mci dose of γ-rays induced typical apoptotic features, such as apoptotic bodies, characteristic sub-G1 peak and so on. More than 6.645 mci of gamma rays cause cell necrosis. Conclusion γ-ray-induced apoptosis of cholangiocarcinoma cells may be one of the important mechanisms of radiotherapy for cholangiocarcinoma. The apoptosis of cholangiocarcinoma cells induced by γ-rays released by 103Pd is closely related to the irradiation dose.
