[目的]分析浙贝母花及其花粉中E族维生素的组成和含量,为浙贝母花资源的开发提供一定的科学依据。[方法]采用高效液相色谱—荧光分析法(HPLC-FLD)检测E族维生素的成分,检测条件如下:Agilent Zorbax RX-SIL(4.6 mm×150 mm,5μm)色谱柱,4.6mm×10 mm保护柱,含0.2%异丙醇和0.8%冰乙酸的正己烷溶液为流动相(1∶4,V/V),流速为1.0 ml/min进行洗脱,柱温25℃,荧光最大激发波长292 nm,最大发射波长328 nm。[结果]首次建立了可同时测定8种维生素E成分的高效液相色谱—荧光分析法,8种成分在检测范围内均具有良好的线性关系,R2>0.999 0,加样回收率大于98%。结果表明浙贝母花中共含有7种E族维生素成分,其中D-α-生育三烯酚含量最高,达181.606 mg/kg,而浙贝母花花粉中E族维生素成分的含量则偏低。[结论]浙贝母花和花粉中均含有较多的E族维生素成分,其中浙贝母花中所含的种类和数量多于花粉,可作为新的药食两用资源进一步开发。同时,本研究建立的HPLC-FLD方法快速高效,适于分析和测定E族维生素的含量。
[Objective] To analyze the composition and content of E vitamins in Fritillaria ussuriensis and its pollen, and provide scientific basis for the development of Fritillaria ussuriensis. [Method] The components of E vitamins were detected by HPLC-FLD with the following conditions: 4.6 mm × 150 mm, 5 μm column, Agilent Zorbax RX-SIL The protective column, n-hexane solution containing 0.2% isopropanol and 0.8% glacial acetic acid as the mobile phase (1: 4, V / V) at a flow rate of 1.0 ml / min. The column temperature was 25 ℃ and the maximum fluorescence excitation wavelength was 292 nm, the maximum emission wavelength is 328 nm. [Result] The HPLC assay - fluorescence analysis method was established for the first time to determine 8 kinds of vitamin E components simultaneously. All the 8 components showed a good linear relationship in the detection range. R2> 0.999 0 and the recoveries were over 98% . The results showed that the Fritillaria cirrhosa contained seven kinds of E vitamins, of which the content of D-α-tocotrienol was the highest (181.606 mg / kg), while the content of E vitamins in pollen of Fritillaria thunbergii was lower. [Conclusion] There were more E vitamins in both Fritillaria and Pollen of Fritillaria cirrhosa, of which Fritillaria thunbergii was more abundant than pollen and could be used as a new edible and dual-purpose resource for further development. At the same time, HPLC-FLD method established in this study is fast and efficient, suitable for analysis and determination of vitamin E content.
