在百合(Lilium)杂交育种中,常采用多个父本混合花粉与一个母本杂交的方法来提高育种效率,为此,研究明确快速父本鉴定方法极为重要。采用SRAP分子标记技术对亚洲百合Red Latin为母本,亚洲百合Matrix和野生种山丹为父本混合授粉后百合杂交Fl代进行父本鉴定。引物筛选共获得4对条带清晰、多态性高的引物(Me1Em4,Me5Em9,Me7Em2和Me7Em7)用于后续SRAP分子标记分析。采用聚丙烯酰胺凝胶电泳技术,对11个F1后代单株和3个亲本材料的SRAP分子标记结果表明:用Me1Em4引物在11个F1后代单株里检测出3条父本特征条带,可以确定8个后代的父本是Matrix,2个后代的父本是山丹;用Me5Em9引物检测出2条父本特征条带,可以确定3个后代的父本是Matrix,1个后代是山丹;用Me7Em2引物检测出2条父本特征条带,可以确定5个后代的父本是Matrix,2个后代是山丹;用Me7Em7引物检测出2条父本特征条带,可以确定3个后代的父本是Matrix。综合分子标记鉴定结果,确定9个株系是Red Latin×Matrix后代,2个株系是Red Latin×山丹的后代,同时表明SRAP分子标记技术可以用于混合授粉后代父本的鉴定。
In Lilium hybrid breeding, the method of multiple male parent pollen hybridization with one female parent is often used to improve the breeding efficiency. Therefore, it is very important to study the method of rapid male parent identification. SRAP molecular markers were used to identify the F1 generation of F1 hybrids of Lily Red Latin as female parent, Asian Lily Matrix and wild species Shandan as male parents. Four primer pairs (Me1Em4, Me5Em9, Me7Em2 and Me7Em7) with clear bands and high polymorphism were obtained by primer screening for subsequent SRAP molecular marker analysis. Using polyacrylamide gel electrophoresis, SRAP molecular markers of 11 F1 progenies and 3 parents showed that the three male-sterile bands were detected in 11 F1 progenies with Me1Em4 primers, The parents of eight offspring were identified as Matrix, the parents of the two offspring were Shandan, and two paternal characteristic bands were detected by Me5Em9 primer. The three offspring parents were identified as Matrix and the other one was Shadan ; Using the Me7Em2 primer to detect the two male-derived bands, it can be determined that the five offspring’s parents are Matrix and the two offspring are Shandan. Two paternal characteristic bands are detected by Me7Em7 primer, and three offspring can be identified The parent is Matrix. According to the results of molecular marker identification, 9 lines were identified as Red Latin × Matrix progeny. Two lines were the descendants of Red Latin × shandan. At the same time, SRAP molecular markers could be used to identify the male parent of mixed pollination.
