目的:探讨二甲基亚砜(dimethyl sulfoxide,DMSO)对多发性骨髓瘤细胞株U266的影响。方法:分别设置含不同终体积分数DMSO的细胞培养体系及空白对照,利用细胞增殖-毒性检测试剂盒(Cell Counting Kit-8,CCK-8)检测不同体积分数的DMSO作用于U266细胞后对其增殖抑制作用,同时利用HE染色观察细胞的形态学变化,探讨其对细胞凋亡的影响。结果:当DMSO体积分数≤0.5%时,U266细胞的生长不受抑制,与空白对照组的抑制率相比,差异无统计学意义;当 DMSO体积分数≥0.6%时,随着体积分数的增加,DMSO对 U266细胞的增殖抑制作用逐渐增强,呈浓度依赖性(P<0.05),并且HE染色结果显示随着DMSO体积分数的增加,凋亡细胞数量逐渐增多。结论:DMSO体积分数≤0.5%时,对U266细胞的生长基本无影响,当体积分数≥0.6%,DMSO对U266具有增殖抑制作用,其中促进细胞凋亡可能是其机制之一。“,”[ABSTRACT]Objective: To explor the effects of DMSO on proliferation of multiple myeloma cell line U266. Methods: Cel Counting Kit-8(CCK-8) was usedto detect the effects of different concentrations of DMSO on proliferation of U266 cell lines for 24h. HE staining was used to observe morphological changes of the cels, and to explore its effect on apoptosis. RESULTS: Compared with the control group, when DMSO≤0.5%,the cel viability rate of treatment group have no difference .And when DMSO≥0.6%,the proliferation inhibition of DMSO on U266 cels gradualy strengthened with increasing of DMSO concentrations. Apoptosis was confirmed by nuclear condensation,membrane blebbing, and formation of round apoptotic bodies. CONLUSION:when DMSO≤0.5%,the DMSO have no effect on U266 cels almostly. when DMSO≥0.6%,the DMSO can inhibit the proliferation of U266 cells and induce apoptosis.
