本工作观察了4个人精子单克隆抗体(BAS_(1—4))对人精子穿透无明带田鼠卵的影响。取获能人精子加入不同稀释度的人精子单克隆抗体处理30min,再加入无透明带的田鼠卵15—20枚,经孵育3h后,观察精子穿卵情况。空白对照组和血清对照组分别用BWW液和灭活小鼠血清代替人精子单克隆抗体。用统计学处理比较实验组和血清对照组受精率及受精指数的差异。结果表明,4个人精子单克隆抗体中,除BAS_4对精子穿卵无抑制作用外,其余3个均有明显抑制作用。以上结果表明,我们所制备的人精子单克隆抗体不仅具有免疫学活性,也具有生物学活性。我们的初步结果为利用人精子单克隆抗体纯化人精子膜抗原,研究精子结构与功能的关系,以及发展一种可能用于局部的新型避孕方法或免疫避孕提供了基础。
In this work, the effects of 4 human sperm monoclonal antibodies (BAS_ (1-4)) on the penetration of human sperm into the voles of Myvate voles were observed. Human sperm were obtained by adding human sperm to different dilutions of human sperm monoclonal antibody for 30 min, and then 15-20 mice with no zona pellucida were added. After incubation for 3 hours, sperm penetration was observed. Blank control group and serum control group were used BWW solution and inactivated mouse serum instead of human sperm monoclonal antibody. Differences in fertilization rate and fertilization index were compared between the experimental group and the serum control group by statistical analysis. The results showed that in addition to BAS_4, no effect was found in the sperm motility of 4 individuals. All three showed significant inhibitory effects. The above results show that we prepared human sperm monoclonal antibody not only has immunological activity, but also has biological activity. Our initial results provide a basis for the purification of human sperm membrane antigens using human sperm monoclonal antibodies, the study of the relationship between sperm structure and function, and the development of a novel contraceptive method that may be used locally or for immunocontraception.
