目的 探讨染矽尘小鼠肺组织转化生长因子 β1(TGF β1)基因的表达在矽肺发病中的意义。方法 采用暴露式气管内注入法给小鼠一次性染矽尘 (0 .2g/kg体重 ) ,染尘后第 1、3、5、7、14、2 8天 ,每组各处死 8只小鼠 ,取肺组织 ,用逆转录聚合酶链式反应 (RT PCR)方法测定小鼠肺组织TGF β1基因的表达。结果 染尘后第 3天 ,小鼠肺组织中TGF β1基因表达开始升高 (1.2 0± 0 .15 ) ;第 7天达高峰 (1.74± 0 .19) ,与对照组 (1.0 1± 0 .16 )和染尘第 5天 (1.35± 0 .15 )比较 ,差异均有显著性 (P <0 .0 5 ) ;第 14天开始下降 ,但直到第 2 8天仍高于对照组。染矽尘小鼠肺组织羟脯氨酸含量随染毒后时间的延长 (1~ 2 8d)呈现持续上升的趋势。结论 TGF β1在矽肺早期的发生、发展过程中可能有重要作用。
Objective To investigate the significance of transforming growth factor β1 (TGFβ1) gene expression in silicosis in silicosis mice. Methods Exposed endotracheal intratracheal instillation of silica dust (0. 2g / kg body weight) to mice once, 1, 3, 5, 7, 14 and 28 days after exposure to dust. The lungs of mice were sacrificed and the expression of TGF β1 gene in lung tissues of mice was measured by reverse transcription polymerase chain reaction (RT PCR). Results On the 3rd day after infection, the expression of TGF β1 gene in the lung tissue of mice began to increase (1.2 ± 0.15), reached the peak on the 7th day (1.74 ± 0.19), compared with the control group (1.0 1 ± 0 .16) and the fifth day of dust (1.35 ± 0.15), the difference was significant (P <0.05); the first 14 days began to decline, but still higher than the control group until the 28th day. The content of hydroxyproline in the lung tissue of the mice exposed to silica dust showed a continuous increasing trend with the prolongation of exposure time (1-28 days). Conclusion TGF β1 may play an important role in the early stage of silicosis.
