目的:通过观察左归丸含药血清对破骨细胞(OC)骨吸收功能的影响,以及成骨细胞(OB)对其的介导作用,探讨其治疗骨质疏松症的作用机理。方法:体外分离培养大鼠OB与OC,观察左归丸含药血清对OC所致骨陷窝数目及面积的影响,以及OB在此过程中的作用。结果:OC与骨片共同培养的第7天,OVX血清能使OC所致的吸收陷窝数目和面积明显增加;而OVX含药大鼠血清能使OC活性明显下降。当OC与OB、骨片共同培养7d后,正常大鼠血清对OC单独培养和OC与OB共同培养所形成骨吸收陷窝的数目和面积没有明显的差异,但共同培养有增加的趋势。而在OVX血清的作用下,OC与OB共同培养所形成的骨吸收陷窝与OC单独培养相比,前者显著高于后者。本实验还发现,OVX含药血清与OB、OC共同培养的骨陷窝数目及面积明显低于OVX含药血清与OC单独培养。结论:在去势状态下,左归丸含药血清不仅对OC有直接的抑制作用,且可通过OB间接对OC产生抑制作用,这可能是其治疗骨质疏松症的作用机理。
Objective: To observe the effect of Zuogui Pill-containing serum on osteoclast (OC) bone resorption function and osteoblast (OB) mediating its effect on the mechanism of osteoporosis treatment. METHODS: Rat OB and OC were isolated and cultured in vitro to observe the effect of Zuogui Pill-containing serum on the number and area of bone lacunae induced by OC, and the role of OB in this process. RESULTS: On the 7th day after co-culture of OC and bone chips, OVX serum could increase the number and area of absorption lacunae caused by OC. However, OVX-containing rat serum could significantly decrease OC activity. When OC was incubated with OB and osteoblasts for 7 days, there was no significant difference in the number and area of bone resorption lacunae formed by OC and OB cultures in normal rat serum, but there was an increasing trend in co-cultivation. Under the effect of OVX serum, the bone resorption lacuna formed by co-culture of OC and OB was significantly higher than that of OC alone. The experiment also found that the number and area of bone lacunae co-cultured with OVX-containing serum and OB, OC were significantly lower than that of OVX-containing serum and OC alone. Conclusion: In the state of castration, Zuogui Pill-containing serum not only has a direct inhibitory effect on OC, but also can inhibit OC by indirect OB, which may be the mechanism of its treatment of osteoporosis.
